ABSTRACT: The mechanistic target of rapamycin (mTOR) is pivotal in programming cellular lipid homeostasis for the structural and functional integrity of cells. Dysregulation of mTOR and consequent altered lipid metabolism are common features in various diseases, including cancers, making mTOR signaling a promising therapeutic target. Therefore, understanding how mTOR activation and inhibition reprogram cellular lipid homeostasis is of physiological and pathological significance. Using mTOR inhibitors in A375 melanoma cells, we performed untargeted lipid profiling to assess changes in lipid composition following mTOR inhibition.
