Project description:Purpose: The State of Rio Grande do Sul is the largest producer of peaches from Brazil. However, it still has low values of productivity when compared to other States. One of the problems associated to it this is the occurrence of drainage soils problems, which can suffer flooding situations potentially hampering the development and productivity such culture. For studies to assist in the selection of flood tolerant genotypes, it is essential to understand the physiological and molecular changes of the plants in situations of oxygen deprivation. Using Illumina Hiseq2500 we performed transcriptome analysis of leaves from ‘Capdeboscq’ (Prunus persica) and ‘Julior’ (Prunus insititia x Prunus domestica) rootstocks under flooding for 48 hours. Methods: The mRNA of Prunus spp. plants cv. Capdeboscq e Julior was generated using deep sequencing, in triplicate, using Illumina Hi-Seq 2500, for the following treatments:I) control: plants received irrigation daily until field capacity; and II) plants exposed to flood stress, maintaining a water level of approximately 3 cm above the ground. The sequence reads that passed quality filters were analyzed at the transcript level using this method: Mapping using STAR and identification of differentially expressed genes (DEGs) was performed with the edgeR (false discovery rates - FDRs of <0.05). RT–qPCR validation was performed using SYBR Green assays. Results: Flooding stress causes important high transcriptional changes in the ‘Capdeboscq’ compared to 'Julior' and this is mainly due to their sensitivity/tolerance levels. ‘Capdeboscq’ had photosynthesis as the most affected physiological process at the molecular level, showing a large number of down-regulated enriched GOs, even though it activated cellular signaling pathways under flooding. 'Julior' was more efficient in defense responses, which include the activation of flavonoid biosynthesis pathways. Conclusions: The analysis of two Prunus spp. rootstocks contrasting to the level of tolerance / sensitivity provide new insights into the process of plant flood stress tolerance.

Project description:Many viruses potently inhibit host protein synthesis while employing unconventional strategies to sustain their own translation, but how and why certain cellular mRNAs continue to be translated remains unclear. Here, we show that during shutoff by Vaccinia Virus (VacV) several host mRNAs increase in polysome occupancy but few, topped by JUN, result in increased protein abundance. Translation of viral mRNAs depended on the small ribosomal protein, Receptor for Activated C Kinase 1 (RACK1) and the eukaryotic Initiation Factor, eIF3. Cryo-electron microscopy (cryo-EM) further showed that eIF3 bound to virus-modified RACK1-containing 40S subunits that exhibit altered head rotation during infection. By contrast, RACK1 and eIF3 were dispensable for JUN translation. Moreover, structurally distinct 5’ untranslated regions (5’UTRs) in viral versus JUN mRNAs conferred differential eIF3 dependencies. Altogether, we reveal how distinct modes of non-canonical initiation support the production of both host and viral proteins that facilitate poxvirus replication despite infection-induced shutoff.

Project description:Arabidopsis does not support the growth and asexual reproduction of the barley pathogen, Blumeria graminis f. sp. hordei Bgh). A majority of germlings fail to penetrate the epidermal cell wall and papillae. To gain additional insight into this interaction, we determined whether the salicylic acid (SA) or jasmonate (JA)/ethylene (ET) defence pathways played a role in blocking barley powdery mildew infections. Only the eds1 mutant and NahG transgenics supported a modest increase in penetration success by the barley powdery mildew. We also compared the global gene expression patterns of Arabidopsis inoculated with the non-host barley powdery mildew to those inoculated with a virulent, host powdery mildew, Erysiphe cichoracearum. Genes repressed by inoculations with non-host and host powdery mildews relative to non-inoculated control plants accounted for two-thirds of the differentially expressed genes. A majority of these genes encoded components of photosynthesis and general metabolism. Consistent with this observation, Arabidopsis growth was inhibited following inoculation with Bgh, suggesting a shift in resource allocation from growth to defence. A number of defence-associated genes were induced during both interactions. These genes likely are components of basal defence responses, which do not effectively block host powdery mildew infections. In addition, genes encoding defensins, anti-microbial peptides whose expression is under the control of the JA/ET signalling pathway, were induced exclusively by non-host pathogens. Ectopic activation of JA/ET signalling protected Arabidopsis against two biotrophic host pathogens. Taken together, these data suggest that biotrophic host pathogens must either suppress or fail to elicit the JA/ET signal transduction pathway. Project description:To reveal dynamic regulation of host gene expression and an overview of events in the infection cycle after SpltNPV infection To analyze cellular responses to SpltNPV infection, a S.litura cell line (SL221) was infected with wild-type baculovirus SpltNPV at a MOI of 1. RNAs were isolated at three time points through the infection cycle (6hpi, 12hpi and 18hpi) and also from control (mock infected) cells. The obtained cellular ranscriptome from the host S.litura was set for illumina strand specific RNA sequencing (RNA-seq) to analyze changes in host cell gene expression upon SpltNPV infection.

Project description:The Aphrophoridae family contains important vectors of Xylella fastidiosa, a serious bacterial plant disease. In olive orchards, nymphs usually feed on the ground-cover vegetation. However, detailed information about their populations and host/non-host plants in some regions threatened by Xylella, such as the northeast of Portugal, is very limited. The goal of our work was to identify the vector species, nymphal development period, and their host and non-host herbaceous plants in olive orchards from northeastern Portugal. Ground-cover plant species hosting or not hosting nymphs were identified during the spring of 2017 to 2019 in olive orchards. Nymphal development period, nymph aggregation, and nymph's preferred feeding height of the ground-cover plants were recorded. The most abundant Aphrophoridae species was Philaenus spumarius followed by Neophilaenus sp. Nymphs developed from April to early May and showed a low number of individuals per foam (generally between one and three). They preferred the middle part of the plants. Philaenus spumarius feeds preferentially on Asteraceae and Fabaceae, and Neophilaenus sp. on Poaceae. Some abundant plants, such as Bromus diandrus, Astragalus pelecinus, Chrysanthemum segetum, Trifolium spp., Caryophyllaceae, and Brassicaceae, were barely colonized by Aphrophoridae nymphs. This knowledge is essential for the selection of the species composition of ground-cover vegetation to minimize the presence of vectors of X. fastidiosa in olive groves.
