Dietary Supplementation with Fermented Chinese Herbal Compounds Enhance Lamb Growth Performance via Targeted Modulation of Gastrointestinal Microbiota

ABSTRACT:

This study adopted the MRM wide-targeted metabolomics approach to detect metabolites in herbal plant samples. Using ultra-high performance liquid chromatography coupled with triple quadrupole mass spectrometry (QTrap 6500+), combined with specific extraction and separation methods, qualitative and quantitative analysis of multiple metabolites in plant samples was achieved, providing data support for the study of metabolic characteristics of herbal plants.

Project description:Purpose: To search candidate synergized compounds from Chinese herbal medicine used in RA therapy Methods: Candidate synergized compounds were searched based on multi-information integrative approaches. In vivo experiment in collagen induced arthritis rat was then carried out to evaluate the efficacy and synergism of compounds combination treatment. RNA sequencing and IPA were used to investigate the synergized mechanism of combination treatment. Results: We found a pair of compound combination which could alleviate the symptom of CIA rats. RNA sequencing and IPA analysis revealed that the synergistic mechanism of combination treatment was related to several canonical signaling pathways and key targets LTA, CD83 and SREBF1. Conclusion: A pair of compound combination has been found. Estrogenic activity of fermented soymilk extracts and soy compounds

Project description:To understand the complex mechanism of estrogen action, multiple assays based on different biological principles is important. Here, multiple assays based on cell, protein and transcription assays were used to evaluate estrogenic activity of soymilk extracts before and after fermentation, and four soy compounds, genistein and daidzein, and their glycosides, genistin and daidzin, respectively. The data obtained by a transcription assay, RNA-sequencing (RNA-seq), was further examined by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) database analysis to understand cell functions, i.e. biological processes and metabolic pathways. Among the cell functions identified, the sets of genes for cell division/cell cycle and DNA replication/DNA repair are significantly up-regulated, whereas those for autophagy are significantly down-regulated, which explains well the enhanced activity of cell proliferation by these materials. The applications of estrogenic activity of soymilk extracts and soy flavonoids, and the contribution of fermentation, include pharmacological benefits, such as bone protection/bone regeneration, cancer chemoprevention, neuroprotection and the treatment of menopausal syndromes.

Gene expression profiling of dendritic cells in different physiological stages under Cordyceps sinensis treatment

Project description:Cordyceps sinensis (CS) has been commonly used as a herbal medicine as well as a health supplement in China for over two thousand years. Although previous studies demonstrated that CS has benefit in immunoregulation and anti-inflammation, the precise mechanism by which CS affects immunomodulation is still unclear. In this study, we adopted duplicate sets of loop-design microarray experiments to examine two different batches of CS and analyzed the effects of CS on dendritic cells (DCs) in different physiology stages, naïve stage and inflammatory stage. We used duplicate sets of loop-design microarray in this study. Each loop experiment consisted of 4 RNA samples, including untreated samples, CS 1μg/ml, LPS 1μg/ml, and LPS 1μg/ml+ CS 1μg/ml.
