ABSTRACT:

The salinity of aquaculture wastewater varies considerably across farming systems, limiting the applicability of conventional Chlorella strains. To improve salt tolerance and broaden application potential, atmospheric and room temperature plasma (ARTP) mutagenesis was employed.

Molecular Adaptation to Ambient Salinity Oscillations in Greater Amberjack (Seriola dumerili): Implications to Aquaculture

Project description:Ambient salinity is one of the crucial abiotic factors that poses substantial impacts on fish growth, development and reproduction. Greater amberjack (Seriola dumerili) is of high economic value because, and its reproduction and survival are sensitive to water salinity. To better understand the molecular adaptive mechanism to salinity fluctuations in greater amberjack, we performed comparative transcriptome analysis for gill and kidney between the optimum salinity (30 ppt, CK) and undesired regimes (10 and 40 ppt). For the gill, the skeletal development was provoked upon either hypo- or hyper-salinity stimuli, and the development of pronephros, as well as vascular endothelial cells and cortisol-mediated mitochondria-rich cell, was activated in response to the salinity alterations in kidney. These enhancements may encourage the maintenance of the gill and kidney structures and alleviate the salinity-induced damage. Ion channels NKCC1 and CFTR and the transporters for ammonium and other substances were highly upregulated in the gills and kidney, respectively, which act important roles in the osmoregulation of greater amberjack. More important, undesirable alterations of ambient salinity were found to pose adverse impacts on the immune function of greater amberjack, which may increase the risk of pathogen infection and reduce the security and yield of aquaculture of greater amberjack. In addition, deviation from the optimum salinity condition may result in undesirable uptake and accumulation of environmental toxins in greater amberjack, which attracts further attention to the food safety. Collectively, these novel findings advance our knowledge on adaptative mechanisms to ambient salinity oscillations in greater amberjack and provide a theoretical guidance for the optimal breeding mode for the aquaculture of greater amberjack.

Atmospheric and Room Temperature Plasma (ARTP) Mutagenesis Improved the Anti-MRSA Activity of Brevibacillus sp. SPR20

Project description:Brevibacillus sp. SPR20 produced potentially antibacterial substances against MRSA. The synthe-sis of these substances is controlled by their biosynthetic gene clusters. Several mutagenesis methods are used to overcome the restriction of gene regulations when genetic information is ab-sent. Atmospheric and room temperature plasma (ARTP) is a powerful technique to make random mutagenesis for microbial strain improvement. This study utilized an argon-based ARTP to conduct the mutations on SPR20. The positive mutants of 40% occurred. The M27 mutant exhib-ited an increase in anti-MRSA activity when compared to the wild-type, by which the MIC values were 250500 and 500 g/mL, respectively. M27 had genetic stability because it exhibited con-stant activity throughout 15 generations. This mutant had similar morphology and antibiotic susceptibility to the wild-type. Comparative proteomic analysis identified some specific proteins that were upregulated in M27. These proteins were involved in the metabolism of amino acids, cell structure and movement, and catalytic enzymes. These might result in the enhancement of the anti-MRSA activity of the ARTP-treated SPR20 mutant. This study supports the ARTP technology to increase the production of valuable antibacterial agents.

Regulation of the anti-immune response by Chlorella intake during swimming stress in mice

Project description:Chlorella has been used as a food supplement and has been reported to have health benefits. We used the forced swimming test to investigate the influence of Chlorella intake during muscle stress training in mice. Swimming time was about two-fold longer for the Chlorella intake mice than for the control mice. Microarray analysis revealed that the global expression profile of muscle from the Chlorella intake mice was similar to that for intact (nonswimming) mice rather than to that for control swimming mice. By contrast, oxidoreductase activity and the leukotriene synthesis pathway were repressed in Chlorella intake mice. According to measurement of the cytokine activities of primary cultures of spleen, Th1 cytokines, such as interferon-gM-cM-^CM-;and interleukin-2, and granulocyte colony-stimulating factors, such as MIP-1aM-cM-^CM-; GMCSF, and IP-10, were decreased in control swimming mice, but these cytokines were rescued in the Chlorella intake mice. We suggest that the immune response during muscle training can be repressed by intake of Chlorella. The intact (nonswimming) and control (swimming) groups were fed normal food, and the Chlorella intake (swimming) group was fed food including 0.5% Chlorella powder. After 14days, the total RNA was isolated from muscle and samples was pooled for each group (n = 10 for each group).

Chlorella ingestion and suppression of resistin gene expression in borderline diabetics: a randomized, placebo-controlled study

Project description:Diabetes and Arteriosclerosis progression are frequently observed in borderline Type 2 diabetes cases. Onset of complications (arteriosclerosis and renal damage) due to Type 2 diabetes is well documented; it is extremely important to prevent or delay their progression. Type 2 diabetes onset and progression has been controlled through dietary habits and exercise, although these remain insufficient. Chlorella ingestion improves blood glucose and cholesterol concentrations in mice and humans, although no reports have evaluated Chlorella effects in borderline diabetics. Therefore, we conducted a randomized, placebo-controlled trial for borderline diabetics using laboratory results and comprehensive gene analysis as outcomes. Chlorella ingestion suppressed resistin gene expression, suggesting that Chlorella may be useful for preventing diabetes onset and ameliorating arteriosclerosis. Subjects were randomly divided into two groups: Chlorella group (n = 28) ingesting Chlorella powder (8.0 g/day) and placebo group (n = 29) ingesting lactose formulation (8.0 g/day) for 12 weeks. Blood and urine were collected every 4 weeks for laboratory tests. Gene expression analysis used RNA extracted from peripheral blood samples before and after 12 weeks of Chlorella or lactose ingestion.

Cross-species dissection of saline-related genes by genetically deciphering a euryhaline microalga Chlorella sp.

Project description:<p>MEM25 and FACHB-9 respectively represent salt-tolerant and freshwater species of the genus Chlorella. By setting salinity of 35 and 105 as the high and low salinity conditions for MEM25, and salinity of 0 and 35 as the high and low salinity conditions for FACHB-9, we measured the metabolome data. To seek the difference in salt tolerance between two classic chlorella species. The measurement conditions are publicized as follows. Approximately 500 mg of algal biomass was collected, followed by metabolite extraction.   Algal cells were suspended in 1 mL mixture of methanol and water (7/3, vol/vol) and kept at − 80 °C for 2 min.   The internal standard used was 2-chloro-l-phenylalanine (0.3 mg mL-1).   The mixture was vortexed at 60 Hz for 2 min, followed by ultrasonication at ambient temperature for 30 min.   Samples were then centrifuged at 13,000 rpm, 4 °C for 15 min.   The supernatants were collected using crystal syringes, filtered through 0.22 μm microfilters and transferred to LC vials, followed by storage at − 80 °C.   LC–MS/MS analyses were performed using an UHPLC system (1290,Agilent Technologies).</p>
