Discovery of novel metabolic pathways and regulatory mechanisms of ammonia-oxidizing bacteria in organic materials stabilization

Project description:Background Ammonia-oxidizing bacteria (AOB) play a crucial role in the microbiome of the global nitrogen cycle. Yet, their regulatory functions in the metabolic pathways involved in organic material stabilization (OMS) are not fully understood. We employed metabolomics, microbiomics, and molecular biology techniques to investigate for the first time the metabolic changes and regulatory mechanisms of OMS in a cow manure and straw biostabilization system inoculated with a novel exogenous consortium-AOB (C-AOB). Results C-AOB engaged in nitrogen metabolism during the thermophilic phase and participated in carbon metabolism during the stabilization phase of OMS. Glutamate was a key metabolite bridging the carbon and nitrogen metabolic networks, which increased during humic formation. C-AOB inoculation stimulated NO2- production, enhanced glycolysis and the tricarboxylic acid cycle, thereby facilitating glutamate conversion and contributing to humic formation. Additionally, NO2- served as a nitrogen source for microbial proliferation, thus enhancing microbial community activity and stability. The observed increase in the abundance of amoA, that encodes ammonia monooxygenase, indicates a close association between C-AOB inoculation and increased glutamate content. Conclusions The key metabolites, enzymes, and genes, particularly those associated with glutamate, played a role in regulating OMS by C-AOB. These findings underscore the regulatory roles of AOB in the metabolic pathways involved in OMS, which is of global concern.

A Bidirectional Regulatory ACLY-NLRP3 Axis Integrates Metabolic and Inflammatory Signaling to Control NLRP3 Inflammasome Activation

Project description:The NLRP3 inflammasome is critical for host defense against bacterial infection and is involved in the pathogenesis of various inflammatory diseases. The aggregation of its sensor protein NLRP3 is a pivotal checkpoint for inflammasome activation. However, the regulatory mechanism of NLRP3 aggregation is not completely understood. Here, we identify the metabolic enzyme ATP-citrate lyase (ACLY) as a direct binding partner that constrains NLRP3 aggregation, membrane translocation and activation. We demonstrate that ACLY interacts with the NLRP3-PYD independently of its enzymatic function, thereby sterically hindering NLRP3 aggregation and suppressing inflammasome assembly. Paradoxically, we find that NLRP3 binding to ACLY inhibits ACLY’s enzymatic activity and remodels cellular metabolism, which counteracts the stimulatory effect of ACLY on NLRP3 inflammasome activation. This reciprocal interaction reveals an essential role of ACLY as a brake for NLRP3 aggregation, and also establishes NLRP3 as a metabolic checkpoint to regulate ACLY-mediated metabolism. Furthermore, we develop a peptide that disrupts the ACLY-NLRP3 interaction and promotes NLRP3 inflammasome activation, which enhances host defense against bacterial infection. Our work delineates a self-limiting circuit for immunometabolic crosstalk, positioning the ACLY-NLRP3 axis as a new therapeutic target for inflammasome-driven pathologies.

Artemisiae Scopariae Herba (Yinchen) suppresses ferroptosis in osteoporosis mice via the Nrf2/SLC7A11/GPX4 pathway

Project description:With the aging of the population, the incidence of postmenopausal osteoporosis (PMOP) is increasing. Artemisiae Scopariae Herba (Yinchen, YC) extracts have been reported to promote osteogenic differentiation and bone formation, but the specific mechanism is still unclear. Aim of the study: To investigate the effect and mechanism of YC on PMOP. Methods: Ultra-performance liquid chromatography-tandem mass spectrometry was used to determine the main potential mechanism of YC, and an ovariectomized (OVX) mouse model was established to evaluate the effects of YC on PMOP and to explore its potential mechanisms. First, the therapeutic effect of YC on PMOP was assessed by CT bone analysis, pathological observation and ELISA detection. Combined with serum ELISA, qRT-PCR and immunohistochemical staining, the potential key anti-PMOP pathway of YC was further explored.

Those bacteria lack classical type III secretion system, and utilize the pore-forming toxin for infection. Two clones CLJ1 and CLJ3 belonging to these taxonomic outliers have been isolated from the same patient at two different times during hospitalization. P. aeruginosa CLJ3 displays antibiotic resistance phenotype, while CLJ1 is more cytotoxic on epithelial and endothelial cells.

Glycerophospholipid remodeling is critical for orthoflavivirus infection

Project description:Flavivirus infection is tightly connected to host lipid metabolism. Here, we performed shotgun lipidomics of cells infected with neurotropic Zika, West Nile, and tick-borne encephalitis viruses, as well as dengue and yellow fever virus. Early in infection specific lipids accumulated, e.g., neutral lipids in Zika and some lyso-phospholipids in all infections. Ceramide levels increased following infection with viruses that cause a cytopathic effect. In addition, fatty acid desaturation as well as glycerophospholipid metabolism were significantly altered. Importantly, depletion of enzymes involved in phosphatidylserine metabolism as well as phosphatidylinositol biosynthesis reduced orthoflavivirus titers and cytopathic effects while inhibition of fatty acid monounsaturation only rescued from virus-induced cell death. Interestingly, interfering with ceramide synthesis had opposing effects on virus replication and cytotoxicity depending on the targeted enzyme. Thus, lipid remodeling by orthoflaviviruses includes distinct changes but also common patterns shared by several viruses that are needed for efficient infection and replication.
