We infected A549 cells with HSV-1, and analized its chromatin structure before infection and at 1, 3 and 8 hours post infection. We report both Human-Human contacts and Human-Virus trans-contacts.

Project description:To discover the mechanism by which herpes simplex virus type 1 (HSV-1) drastically reshapes the host cell transcriptional patterns during infection, we performed RNAseq. We infected A549 cells with HSV-1, and analized host and viral expression at 1, 3 and 8 hours post infection. We report both a drastical human transcription shutoff parallel with an increase in viral transcription.

Identification of miR-203 Regulated Genes During Sendai Virus Infection

Project description:The goal of this experiment was to determine gene expression changes during Sendai virus infection as the result of expression or inhibition of miR-203 in A549 cells. The gene expression profiling experiment was performed with 4 groups (mock infected, Sendai virus infected, Sendai virus infeceted in the presence of exogenous miR-203, and Sendai virus infected in the presence of miR-203 inhibitor) with 3 biological replicates for each group. Total RNA was purified from A549 cells that were mock infected or infected with Sendai virus (Cantell strain, 5pfu/cell) alone or in the presence of miR-203 mimic or inhibitor for 10 hours.

Project description:Gene expression profiles of two Pseudomonas aeruginosa taxonomic outlier clinical isolates, CLJ1 and CLJ3 [CLJ3] Pseudomonas aeruginosa taxonomic outliers emerged recently as infectious for humans, provoking hemorrhagic pneumonia. Those bacteria lack classical type III secretion system, and utilize the pore-forming toxin for infection. Two clones CLJ1 and CLJ3 belonging to these taxonomic outliers have been isolated from the same patient at two different times during hospitalization. P. aeruginosa CLJ3 displays antibiotic resistance phenotype, while CLJ1 is more cytotoxic on epithelial and endothelial cells.

Glycerophospholipid remodeling is critical for orthoflavivirus infection

Project description:Flavivirus infection is tightly connected to host lipid metabolism. Here, we performed shotgun lipidomics of cells infected with neurotropic Zika, West Nile, and tick-borne encephalitis viruses, as well as dengue and yellow fever virus. Early in infection specific lipids accumulated, e.g., neutral lipids in Zika and some lyso-phospholipids in all infections. Ceramide levels increased following infection with viruses that cause a cytopathic effect. In addition, fatty acid desaturation as well as glycerophospholipid metabolism were significantly altered. Importantly, depletion of enzymes involved in phosphatidylserine metabolism as well as phosphatidylinositol biosynthesis reduced orthoflavivirus titers and cytopathic effects while inhibition of fatty acid monounsaturation only rescued from virus-induced cell death. Interestingly, interfering with ceramide synthesis had opposing effects on virus replication and cytotoxicity depending on the targeted enzyme. Thus, lipid remodeling by orthoflaviviruses includes distinct changes but also common patterns shared by several viruses that are needed for efficient infection and replication.

Finding the LC3-interactors in senescent human A549 cells during influenza A virus infection

Project description:To screen age-related regulators of mitophagy, we examined LC3-interacting proteins by proximity-dependent biotin identification. LC3B was cloned into a mammalian expression vector tagged with a mitochondrial OMM-targeted APEX2 (mito-APEX) label for proximity-based identification of LC3-interacting proteins localized or trafficked to mitochondria during influenza virus infection. Senescent A549 cells, transfected with the mito-APEX-tagged LC3 plasmid for 18 hours, revealed mitochondrial localization of LC3. Following 18 hpi and biotin affinity capture, biotinylated proteins were purified and analyzed by mass spectrometry to identify LC3-interacting proteins.

Gene expression changes due to influenza virus induced miRNAs

Project description:The goal of this experiment was to determine gene expression changes during influenza A virus infection as the result of expression influenza virus inducible miRNAs in A549 cells. The gene expression profiling experiment was performed with 4 groups (mock infected, influenza A virus infected, influenza A virus infected in the presence of exogenous miR-141, miR-374b, miR-449b, miR-518b, and miR-1263, and influenza A virus infected in the presence of exogenous miR-147b, miR-190b, miR-199a, miR-512-5p, and miR-874 with 3 biological replicates for each group. Total RNA was purified from A549 cells that were mock infected or infected with influenza A virus (A/WSN/33, 5pfu/cell) alone or in the presence of miRNA mimics 10 hours after treatment.
