ABSTRACT: To investigate the metabolic shift mechanism of Komagataeibacter hansenii(K. hansenii)HDM1-3 from the bacterial growth phase to the bacterial cellulose (BC) synthesis phase,untargeted metabolomics analysis was performed on K. hansenii HDM1-3 cultures at the early fermentation stage(18h) and late fermentation stage(36h).Pathway enrichment analysis identified six significantly enriched key metabolic pathways:Starch and sucrose metabolism,Glycerophospholipid metabolism and Galactose metabolism.Differential metabolite screening revealed a total of 211 differential metabolites. Compared to 18h,92 metabolites were upregulated and 119 metabolites were downregulated in 36h.Further screening identified significant abundance differences between 18h and 36h for the following metabolites:UDP-glucose, ADP-glucose andMelibiose.These metabolites serve as potential biomarkers distinguishing different fermentation phases.The research enriches the metabolomic understanding of K. hansenii and provides a theoretical reference for enhancing BC biosynthesis yield in this bacterium in future studies.
