ABSTRACT: ‘Tiegun’ yam (Dioscorea polystachya Turcz.) is regarded as a medicinal and edible crop with extremely health-preserving values. However, the types and accumulation of bioactive substances in ‘Tiegun’ are incompletely elucidated. We firstly explored the bioactive substance accumulation across the process of ‘Tiegun’ tuber growth. Some well-recognized functional substances such as isoflavones and flavanols continuously accumulated significantly in the mature tuber. The flavonoid biosynthesis pathway was analyzed through transcriptome sequencing on the different tuber growth stage. DpERF1-4 expression levels and the contents of ethylene and gibberellin were significantly positively correlated with flavonoid accumulation. Exogenous application of 0.5 mM ethephon at the early stage could increase flavonoid content in tubers by 31-fold, and simultaneously upregulating the expression levels of DpERF1-4 and key flavonoid biosynthesis genes. This is the first report about DpERF response to ethylene signal and its promotion the key flavonoid biosynthesis genes expression.
