Proteomic analysis of osteocyte line (MLO-Y4) with knockout of cystic fibrosis transmembrane conductance regulator (CFTR) or chloride deprivation treatment

ABSTRACT:

Mass spectrometry–based proteomic analysis of an osteocyte line, MLO-Y4, was conducted to profile global protein expression changes under two experimental settings, 1) CRISPR-Cas9-based knockout of CFTR versus wild-type; 2) Cl- deprivation versus normal extracellular medium.

We generated mouse embryonic stem cells (mESC) constitutively depleted of Gal-1 and/or Gal-3 using CRISPR-Cas9, and assessed the transcriptomic effects of the individual or concomitant depletion of Gal-1 and Gal-3 in mESCs differentiated by deprivation of the leukaemia inhibitory factor (LIF) for 7 days. Wild-type cells were employed as control.

The vulnerability of actin cytoskeleton to disulfide stress dictates high SLC7A11-driven cell death upon glucose deprivation

Project description:Solute carrier family 7 member 11 (SLC7A11; also known as xCT) is a cystine transporter frequently overexpressed in cancers. Cancer cells with high expression of SLC7A11 (SLC7A11high) are resistant to ferroptosis but exquisitely sensitive to glucose deprivation-induced cell death, although the underlying mechanism of the latter cell death remains poorly understood. Here we show that We found that glucose deprivation induced a novel type of cell death in SLC7A11high cancer cells that was different from apoptosis or ferroptosis. And the bioorthogonal chemical proteomics analysis revealed that a large amount of actin cytoskeleton proteins were shown with significantly dysregulated disulfides induced by glucose starvation in SLC7A11high cancer cells. In mechanism, SLC7A11high mediated cystine uptake and subsequent reduction to cysteine forced disulfide stress on actin cytoskeleton upon glucose deprivation through draining intracellular reduced nicotinamide adenine dinucleotide phosphate (NADPH) pool, which was independent of generating reactive oxygen species (ROS). The disulfide stress on the actin cytoskeleton induced rapidly contracted actin filaments and detached from the plasma membrane, which incurred cell death. In the end, using whole-genome CRISPR/Cas9 knock-out screening we identified NCKAP1 as a downstream factor of SLC7A11 to promoting cell death upon glucose deprivation by inhibiting branched actin filaments.

Label-free proteomic analysis of neuroglobin-knockout breast cancer cells

Project description:Neuroglobin (NGB) is an oxygen-binding protein that takes part in several processes that safeguard redox homeostasis, gaining an important protective role in neuronal and extra-neuronal cells. NGB remarkably exerts its function upon up-regulation by NGB-inducible molecules such as 17β-estradiol (E2) and H2O2. Despite this, the essential mechanisms related to NGB function remain still undefined. Human MCF-7 breast cancer cells knocked out (KO) for NGB gene obtained using CRISPR/Cas9 technology were analyzed by shotgun label-free quantitative proteomics in comparison with control cells. Differential proteomics experiments were also performed using NGB-KO cells after treatment with E2, H2O2, and E2+H2O2 versus their corresponding non-treated controls.

Proteomic analysis of glutamine deprivation in HepG2 cells

Two independent cultured cell lysates, namely control and glutamine deprivation, were marked with iTRAQ labels and subjected to LC-ESI-MS/MS analysis. The tryptic peptides of two samples were labeled with mass 113 (control) and 119 (glutamine deprivation) isobaric iTRAQ tags.

Effect of sleep deprivation on non-small cell lung cancer metastasis

Project description:Recent research has shed light on the influence of sleep deprivation on tumor progression, particularly in non-small cell lung cancer (NSCLC), although the precise underlying mechanisms remain poorly understood. Several studies have highlighted alterations in immune cell dynamics and intercellular communication as key consequences of sleep deprivation. Moreover, heightened sympathetic nervous system activity has been implicated in this context. To explore these phenomena further, transcriptome sequencing was conducted in tumor-bearing mice subjected to normal sleep conditions versus sleep deprivation. This approach aimed to delineate the molecular mechanisms linking sleep deprivation to NSCLC progression.
