Targeted LC-MS analysis for lipidomics of mice intestinal samples

ABSTRACT:

Mice of C57BL/6 were received a single dose of 12 Gy of the whole abdominal radiation (WART) to establish acute radiation bowel injury model. Approximately 12 hours post-irradiation, mice were orally administrated with PBS, DY802 and TG-CS@DY802 respectively. For 5 days consecutive treatments, mice were suicided and collected with intestinal tissues for Liquid Chromatograph Mass Spectrometer (LC-MS) analysis of lipids and lipid-like molecules.

Project description:Transcriptional profiling of intestinal epithelial cells expressing either Negative, Sublow, Low or High levels of the Sox9-EGFP reporter transgene FACS-isolated from jejunum of non-irradiated mice or at 5 days after 14Gy abdominal irradiation. In both groups, mice were treated for 5 consecutive days with either IGF1 or vehicle via mini-pumps (Alzet 1007D, IGF1 at 10mg/ml) implanted subcutaneously immediately following radiation. 4 distinct cell populations FACS-isolated based on levels of expression of the Sox9-EGFP reporter transgene (Sox9-EGFP Negative, Sublow, Low and High cells). 4 conditions: Non-irradiated/vehicle vs Non-irradiated/IGF1 vs Irradiated/vehicle vs Irradiated/IGF1. Biological replicates: 7 independent non-irradiated mice treated with vehicle - 3 independent non-irradiated mice treated for 5 days with IGF1 - 3 independent irradiated mice studied at day 5 post-irradiation treated with vehicle - 3 independent irradiated mice studied at day 5 post-irradiation treated for 5 days with IGF1.

Matrine derivative 1b(MASM) offers radioprotection and modulates acute lethal total-body irradiation-induced alterations of gene expression

Project description:To further investigate the potential molecular basis of the protective effects of MASM on irradiation (6.5Gy) damage, gene expression analysis was conducted on rats liver tissues using microarrays. Pre-treatment with MASM prevented differential expression of 53% (766 genes) of 1445 genes differentially expressed in response to radiation. Pathway enrichment analysis indicated that these genes were mainly involved in a total of 21 pathways, such as metabolic pathways, pathways in cancer, and MAPK signaling pathway. The rats were randomly assigned to one of the three following treatment groups (10-12 animals per group): normal control, radiation and MASM dose (30 mg/kg body weight/day) + radiation. MASM dissolved in double distilled water were administered intragastrically to the male animals for 3 consecutive days before irradiation. Radiation induced gene expression in rat liver was measured at 24 hours after 6.5 Gy exposure.

Effects of Low-Dose Irradiation Using Unsealed Caesium-137 on Cultured Normal Human Cells

Project description:Caesium-137 (Cs-137) is one of the major radionuclides appearing in the natural environment after nuclear power plant accidents. However, the biological effects of low-dose internal irradiation with this radionuclide remain unclear. We developed an experimental model for studying low-dose internal irradiation using cultured human cells. The cells were incubated in the culture medium supplemented with unsealed Cs-137 chloride. We used the Monte Carlo simulation method for measuring internal irradiation because making direct measurements was not possible. The simulation revealed that 96.40%?99.70% of the internal irradiation involved ?-particles and other electrons. During the experiment, a gradual incorporation of Cs-137 in the cells, and the absorbed dose rate increased in a time-dependent manner. In addition, the number of ?-H2AX and 53BP1 nuclear foci in the cells increased by internal irradiation in a dose-dependent manner. Microarray analysis revealed time-dependent alterations in gene expression caused by the radiation. These results demonstrate that our experimental system can be useful in the investigation of the effects of low-dose internal irradiation.

Transcriptomic profiling of intestinal tissue in a rat model of radiation-induced enteritis treated with Changbing Formula

Project description:Radiation-induced enteritis is a common and severe complication of abdominal radiotherapy, yet effective therapeutic strategies remain limited. In this study, a Sprague–Dawley rat model of radiation-induced intestinal injury was established using a single dose of 10 Gy X-ray irradiation. Rats were divided into control, radiation model, and treatment groups, with the treatment group receiving Changbing Formula intervention following irradiation. Intestinal tissues were collected for transcriptome sequencing to investigate gene expression changes associated with radiation injury and therapeutic intervention. High-throughput RNA sequencing was performed using the Illumina NovaSeq platform, and gene-level expression profiles were generated to elucidate molecular mechanisms underlying radiation-induced enteritis and the protective effects of Changbing Formula.

Sexual dimorphism of gut microbiota dictates therapeutics efficacy of radiation injuries [RNA-seq]

Project description:Accidental or iatrogenic ionizing radiation exposure precipitates acute and chronic radiation injuries. Traditional paradigm of mitigating radiotherapy-associated side effects has ignored the gender-specific dimorphism of patients. Here we examined the effects of sexual dimorphism on therapeutic agent efficiencies in murine models. High-throughput sequencing of host mRNA showed that different treatments reprogrammed the spectrum of mRNA expression in small intestines of male or female mice, respectively. Collectively, our observations demonstrate that therapeutic strategy efficiencies for radiation toxicity might be dependent on the gender of patients. We aim to uncover the underlying mechanisms by which simvastatin or high fat diet fights against radiotherapy-induced gastrointestinal tract toxicity for male or female mice. The mice were exposed to 12 Gy total abdominal irradiation (TAI), then treated with simvastatin for male mice or fed with high fat diet for female mice. The small intestine tissues were extracted from male mice without TAI, exposed to TAI only, and exposed to TAI combined with simvastatin treatment as one cohort, and from female mice without TAI, exposed to TAI only, and exposed to TAI combined with high fat diet feeding as the other cohort.

Genome-wide analysis of the therapeutic effect of NeuroD on irradiation-induced intestinal injury

Project description:The objective was to investigate the therapeutic effect of NeuroD on irradiation-induced intestinal injury at gene expression level. Gene expression profilings between NeuroD and EGFP treated mice which were received 9 Gy total body irradiation were compared. All mice were given 9 Gy radiation and divided into two groups (3 mice per group). After radiation, mice were immediately administered with an intraperitoneal injection of 0.1 mM EGFP or NeuroD-EGFP. EGFP treated mice were served as controls. 12 hours post radiation, mice jejunum tissues were isolated. Total RNA obtained from jejunum tissues and subjected to gene expression profiling assays.
