ABSTRACT: Rheumatoid arthritis (RA) monocytes are primed for inflammatory activation, but their disease-intrinsic molecular features have not been systematically profiled across omics layers. We aimed to define RA-associated alterations in primary human monocytes using an integrated multi-omics approach under controlled conditions. Peripheral blood CD14+ monocytes from RA patients and matched healthy donors were analyzed in the undifferentiated state (M0) or after differentiation into classically activated M(IFNy+LPS) or alternatively activated M(IL-4) macrophages, with or without acute lipopolysaccharide (LPS) stimulation. Untargeted metabolomics (untargeted LC–MS/MS) revealed a strong disease-driven signature, characterized by decreased nucleotides, redox metabolites and carnitines, while amino acids and urea cycle intermediates were increased. This MetaboLights Study contains untargeted LC-MS/MS data of a validation cohort wherein RA vs healthy monocytes were sampled immediately after CD14+ MACS isolation.
