Untargeted metabolomic analysis of Stylosanthes guianensis roots and nodules following Bradyrhizobium inoculation in acidic soil

ABSTRACT: Prior to transplantation into acidic soil, 30-day-old Stylosanthes guianensis seedlings were inoculated with Bradyrhizobium (Br) strain LZ3-2. Sixty days after transplantation, root and nodule samples were collected, flash-frozen in liquid nitrogen, and stored at negative 80 degrees Celsius for subsequent untargeted metabolomic analysis.

Gene Expression in Acute Appendicitis

Project description:Gene expression was evaluated in 9 appendix samples removed from patients who went to the operating room with the diagnosis of acute appendicitis and 4 samples removed for non-inflammatory reasons. A circumferential piece of tissue was obtained at the distal aspect of each specimen. The tissue was flash frozen at -80 degrees for later processing. Frozen specimens were homogenized into TriReagent and RNA was isolated according to the manufacturer's instruction. RNA was processed and hybridized to Affymetrix microarray. Wild-type and ESTAIG1 Col samples were treated with estrogen and flash frozen 0, 60 and 120 minutes later. Each sample type has three biological reps, except ESTHAT22.120, which has two. There are therefore 17 total samples. mRNA samples were sequenced by Otogenetics and then analyzed using the Tuxedo suite of applications.

Expression profiling of F2 progenies from C57BL/6J x DBA/2J

Project description:Expression profiling of a cohort of mice was derived from a standard F2 intercross using two standard inbred strains, C57BL/6J and DBA/2J. In this study, we selected 127 male animals that had been raised on a standard rodent chow diet (Purina Chow from Ralston-Purina Co., St. Louis, MO) for eight weeks and then switched to a high fat diet for sixteen additional weeks. At 20 weeks of age, the mice were euthanized and liver tissues were collected and flash frozen in liquid nitrogen and stored at -80 degrees C prior to RNA isolation. All procedures of housing and treatment of animals were performed in accordance with IACUC regulations.
