LC-MS based metabolomics analysis of mouse fecal metabolites reveals microbiota-derived products that impair colonic stem cell regeneration in Lamtor5+/- mice

ABSTRACT: This study aimed to investigate the metabolic alterations in mouse fecal samples and identify microbiota-derived products responsible for impaired colonic stem cell regeneration in Lamtor5+/- mice. A total of four experimental groups were included: wild-type (WT) mice treated with PBS (control), Lamtor5+/- mice treated with PBS, WT mice treated with DSS, and Lamtor5+/- mice treated with DSS. Fecal samples were collected and analyzed using liquid chromatography-mass spectrometry (LC-MS) in both positive and negative ion modes. Untargeted metabolomics data preprocessing included peak filtering, missing value imputation, normalization, and log transformation. Differential metabolites were identified and annotated using KEGG and HMDB databases. The results provide insights into how the absence of the H gene alters gut microbiota-derived metabolites and subsequently reduces colonic stem cell regeneration capacity.
