Project description:Cellular senescence is a hallmark of aging, and the accumulation of senescent cells (SnCs) accelerates the aging process and contributes to aging-related organ disorders. The PRDMs exhibit robust transcriptional regulatory activity and govern a wide range of biological processes. However, the role of PRDMs in cellular senescence and aging remain unclear. Here, we have demonstrated that PRDM16, among PRDMs, is decreased significantly in multiple organs of aged mice compared to young mice. Global Prdm16 deletion contributes to cellular senescence in various organs, including the kidney, heart, lung, brain hippocampus, stomach, and gut; subsequently leading to accelerated aging-related organ injury. Furthermore, tubular specific Prdm16 deletion aggravates irradiation induced kidney aging, and aging-related kidney disease subjected to ischemia reperfusion surgery. Exogenous PRDM16 gene delivery by lentivirus effectively attenuates cellular senescence in vitro and in vivo. Mechanistically, PRDM16 improves glutathione metabolism and inhibits oxidative DNA damage, the driving force of senescence. Especially, PRDM16 increases glutathione-S-transferase activity by upregulating the transcription of GSTM1. Transfection of GSTM1 restored cellular senescence and kidney aging caused by PRDM16 deficiency. Taken together, we provide a potential target for investigating the anti-aging therapy.

Project description:As the global aging society intensifies, the incidence of thyroid diseases among the elderly is increasing annually, with hypothyroidism being particularly common. Studies have shown that in healthy elderly individuals, thyroid-stimulating hormone (TSH) levels are significantly elevated, while thyroid hormone (TH) levels are notably normal or decreased. However, the molecular mechanisms underlying thyroid aging and function remain unclear. To understand thyroid aging, it is essential to study the molecular and functional characteristics of various thyroid cell types during the aging process. We conducted single-cell sequencing of thyroids from different age groups and identified eight typical cell types and five thyroid epithelial cell subtypes (EPI). During aging, the expression levels of thyroid hormone synthesis-related genes (TSHR, TG, TPO) increased, suggesting that EPI cells adjust their gene expression patterns in response to elevated TSH. With advancing age, rhythmic disruption and cellular senescence signals accumulated in thyroid. Further analysis indicated that the senescent EPI cell subpopulation (CDKN1A_EPI) exhibited functional decline. Additionally, we unexpectedly found that the expression of the core circadian gene BMAL1 (ARNTL) was downregulated in the CDKN1A_EPI cells of elderly individuals. To verify this finding, we used a thyroid-specific knockout mouse model and found that BMAL1 downregulation inhibited NFKBIA expression, exacerbating cellular senescence. Finally, cell line knockout experiments and transcriptome sequencing further confirmed that BMAL1 knockout led to decreased expression of NF-kB Inhibitor Alpha (NFKBIA), a key mechanism driving thyroid cell senescence. This discovery not only reveals the relationship between thyroid cell aging and circadian rhythm disruption but also enriches our understanding of the mechanisms underlying thyroid aging.

Project description:Age-related thymic involution is the hallmark of immune aging, leading to susceptibility to cancers and emerging pathogens such as SARS-Cov2 in the elderly. However, the precise mechanisms underlying the thymic involution and the effect on compromised peripheral T-cell function is unknown. Here by scRNA-seq and TCR-seq, we captured and analyzed cells in the thymus and peripheral blood from the young and elderly. Collectively, our data constitute the first detailed single-cell analysis of ageing-related human thymic involution and T-cell homeostasis in peripheral, providing the insights into human immune aging and potential strategies to rejuvenate the involuted thymus and compromised T-cell immunity.

Project description:Heterogeneous populations of human bone marrow-derived stromal cells (BMSC) are among the most frequently tested cellular therapeutics for treating degenerative and immune disorders, which occur predominantly in the aging population. Currently, it is unclear whether advanced donor age and commonly associated comorbidities affect the properties of ex vivo-expanded BMSCs. Thus, we stratified cells from adult and elderly donors from our biobank (n = 10 and n = 13, mean age 38 and 72 years, respectively) and compared their phenotypic and functional performance, using multiple assays typically employed as minimal criteria for defining multipotent mesenchymal stromal cells (MSCs).We found that BMSCs from both cohorts meet the standard criteria for MSC, exhibiting similar morphology, growth kinetics, gene expression profiles, and pro-angiogenic and immunosuppressive potential and the capacity to differentiate toward adipogenic, chondrogenic, and osteogenic lineages.We found no substantial differences between cells from the adult and elderly cohorts. As positive controls, we studied the impact of in vitro aging and inflammatory cytokine stimulation. Both conditions clearly affected the cellular properties, independent of donor age. We conclude that in vitro aging rather than in vivo donor aging influences BMSC characteristics.

Project description:The rapidly aging population has an increased alcohol use, resulting in increased risk and mortality of alcohol-associated acute pancreatitis in the elderly population. We established an aging- and alcohol-associated acute pancreatitis mouse model and applied spatial transcriptomics to profile heterogenous tissue composition in the mouse pancreas and the associated gene expression. By comparing with the young cohorts, our analysis has revealed the key differentially expressed genes in functional tissue compartments for understanding the mechanisms and developing targeted therapeutic strategies.

Project description:Mammalian aging is characterized by the progressive loss of tissue integrity and function manifesting in ill health and increased risk for developing multiple chronic conditions. Accumulation of senescent cells in aging tissues partly contributes to this decline as targeted depletion of senescent cells in vivo ameliorates many age-related phenotypes. However, the fundamental molecular mechanisms responsible for the decline of cellular health and fitness during senescence and aging are largely unknown. In this study, we investigated whether chromatin-mediated loss of transcriptional fidelity, known to contribute to fitness and survival in yeast and worms, also occurs during human cellular senescence and mouse aging. Our findings reveal that aberrant transcription initiation inside genes is widespread in senescence and aging. It co-occurs with changes in the chromatin landscape and formation of non-canonical transcription start sites. Interventions that alter spurious transcripts have dramatic consequences on cellular health primarily affecting intracellular signal transduction pathways. We propose that spurious transcription is a conserved hallmark of aging that promotes a noisy transcriptome and a degradation of coherent transcriptional networks.
