Global CIL LC–MS Metabolomic Profiling of Kidney Cancer Samples

ABSTRACT: This study presents a comprehensive metabolomic profiling of kidney cancer samples using a chemical isotope labeling (CIL) LC–MS platform. The workflow includes systematic sample preparation, isotope labeling targeting specific metabolite functional groups, and high-resolution liquid chromatography–mass spectrometry (LC–MS) analysis. A pooled quality control (QC) sample, generated by combining aliquots from individual samples, was included to monitor analytical performance and ensure data consistency across the dataset. Metabolite detection and identification were performed using established CIL metabolomics pipelines, enabling enhanced coverage and improved quantification accuracy. Statistical analyses, including principal component analysis (PCA), partial least squares–discriminant analysis (PLS-DA), and volcano plot–based differential analysis, were conducted to evaluate metabolic variation and identify significantly altered metabolites across sample groups.
