Dynamic force sensing by bone delays aging and extends lifespan

ABSTRACT: Aging is accompanied by systemic metabolic remodeling, and exercise-based interventions can delay aging; however, the circulating metabolic mediators remain poorly defined. Dynamic tibial axial loading (DTAL) is a controlled bone mechanosensing intervention that has been shown to delay aging and extend healthspan and lifespan in mice. To characterize its systemic metabolic effects, we performed plasma metabolomic profiling of aged mice subjected to DTAL or no DTAL. Differential metabolite analysis revealed that DTAL significantly elevated geroprotective metabolites, including melatonin, betaine, glutathione, and adenosine triphosphate (ATP), while reducing ozagrel, isatin, thioisonicotinamide, suberylglycine, and indoxyl sulfate. These metabolomic changes suggest that DTAL reshapes circulating metabolism toward an anti-aging state, with increased protective and energy-related metabolites and decreased potentially stress-associated metabolites. This dataset provides a resource for investigating the systemic metabolic mediators of bone mechanosensing-mediated healthspan and lifespan extension, and may aid in identifying exercise-mimetic metabolic signatures for aging interventions.
