Untargeted liver metabolomic profiling of hepatocyte-specific Serpine2-knockout mice following ischemia-reperfusion injury
Ontology highlight
ABSTRACT: Untargeted metabolomics was performed to investigate hepatic metabolic changes associated with hepatocyte-specific Serpine2 deletion after ischemia-reperfusion injury. Eight liver samples were collected from Serpine2-floxed control mice (NC) and hepatocyte-specific Serpine2-knockout mice (HKO), with four biologically independent mice per group. Both groups underwent 90 min of approximately 70% warm hepatic ischemia followed by 6 h of reperfusion. Metabolite extracts were analyzed by UHPLC-MS using a Vanquish Flex system coupled to a Q Exactive HF-X mass spectrometer. Pooled quality-control samples were included, and data were processed using MS-DIAL to compare metabolic profiles between groups.
INSTRUMENT(S): Gas Chromatography MS - positive - - Vanquish Flex UPLC - Q Exactive HF-X (positive), Liquid Chromatography MS - negative - - Vanquish Flex UPLC - Q Exactive HF-X (negative)
