Spatial metabolomics data of seed development in the soybean cultivar QiHuang No. 34. (MS Imaging)

ABSTRACT: Spatial metabolomics profiling was performed on materials collected from four stages of seed development (including the early maturation stage and three time points of the mid-maturation stage) of the soybean cultivar QH34 grown in a field environment. For spatial metabolomics, tissues were cryosectioned at 12 µm on a Leica CM1950 cryostat, thaw-mounted onto pre-chilled ITO conductive glass slides, and vacuum-dried for 30 min. The sections were then sprayed with 15 mg/mL DHB matrix (acetonitrile:water = 90:10) using a TM-Sprayer (60 °C, 0.12 mL/min, 8 psi, 25 cycles, with 5 s of drying between cycles), and acquired on a timsTOF fleX mass spectrometer in DataImaging mode with an imaging resolution of 20 µm and a mass range of m/z 50–1,300. Raw data were imported into SCiLS Lab and normalized by RMS to generate ion images.
