Integrated Metagenomic and Metabolomic Analysis Reveals Microbial Succession and Metabolic Profiles of Huang Suan Co-Fermented with Post-Flowering Young Rapeseed Pods and Allium chrysanthum Regel

ABSTRACT: Huang Suan is a traditional fermented vegetable from Guizhou prepared from young rapeseed pods and Allium chrysanthum. This study integrated metagenomics and metabolomics to investigate the microbial succession and metabolic remodeling during fermentation. Metagenomic analysis revealed that Latilactobacillus sakei, Companilactobacillus alimentarius, and Lactiplantibacillus plantarum became the dominant taxa, accompanied by rapid acidification. The decline of opportunistic pathogens co-occurred with low pH, ethanol stress from the added baijiu, and competitive exclusion. Metabolomic profiling showed significant accumulation of flavonoid aglycones (e.g., quercetin, isorhamnetin) and specific amino acids, alongside enhanced in vitro antioxidant activities. Multi-omics association analyses indicated that dominant LAB were associated with overall metabolite categories, while low-abundance taxa co-varied with specific compounds. Moreover, metagenomic data suggested a genetic potential for cell wall degradation and flavonoid deglycosylation via key CAZymes (e.g., CE8, GH1). These findings provide a descriptive foundation for understanding this novel fermentation system.
