TREM2-Positive Macrophages and Renal Ceramide Dysregulation in Diabetic Kidney Disease
Ontology highlight
ABSTRACT: This study investigated the effects of TREM2 deficiency and high-fat diet/streptozotocin (HFD/STZ)-induced diabetes on renal lipid metabolism in mice. Kidney tissue samples were collected from four experimental groups, with five biological replicates per group (20 samples in total): (A) TREM2 knockout mice maintained on a normal diet, (B) TREM2 knockout mice subjected to HFD/STZ treatment, (C) wild-type control mice maintained on a normal diet, and (D) wild-type mice subjected to HFD/STZ treatment. Untargeted lipidomic profiling was performed using ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry (UHPLC-MS/MS) on a Vanquish UHPLC system and an Orbitrap Exploris 120 mass spectrometer. Data were acquired in both positive and negative ionization modes. A pooled quality control (QC) sample was prepared by combining equal aliquots of experimental samples and analyzed repeatedly to assess analytical stability and reproducibility. Mass spectrometry data were converted to mzXML format using ProteoWizard and processed using XCMS for peak detection, retention time alignment, and peak integration. Lipid annotation was performed using an in-house R-based workflow and the LipidBlast 2022 database. Following data preprocessing, 3,585 annotated lipids were retained for analysis. Multivariate statistical analyses, including principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA), were performed to investigate lipidomic differences between experimental groups. This dataset provides a resource for investigating renal lipid remodeling, particularly alterations in ceramide and sphingolipid metabolism, associated with TREM2 deficiency and diabetic kidney disease.
