THEMIS2 promotes ovarian cancer progression and cisplatin resistance through inflammatory signaling and lipid metabolic reprogramming: an integrated multi-omics and experimental study

ABSTRACT: We conducted an integrated pan-omics analysis of THEMIS2 across six molecular layers from TCGA-OV (mRNA, CNV, mutation, methylation, miRNA, RPPA), four independent GEO cohorts (n=450), DepMap CRISPR essentiality screens, LINCS L1000 drug signatures, single-cell RNA-seq (GSE165897, 51,786 cells), and spatial transcriptomics (GSE211956, 8 Visium samples). Functional validation was performed using THEMIS2 knockdown in A2780 cells with CCK-8, EdU, flow cytometry, and Transwell assays. A three-omics (transcriptomic, proteomic, and metabolomic) integrative analysis was conducted under cisplatin treatment with THEMIS2 knockdown to elucidate downstream mechanisms.
