ABSTRACT: In this experiment a hormonal protocol was applied to control follicle growth to yield larger or smaller preovulatory follicle and CLs and consequently different circulating Progesterone (P4) concentrations during early diestrus. The two different animal's group are: high or low progesterone levels. The effects of these progesterone levels was tested in the ovaries (follicle fluid) of the cow.

Project description:We utilized whole-genome microarrays (Illumina Mouse WG-6) to measure changes in mRNA abundance levels induced by steroid hormones in primary neurons and astrocytes. To stimulate gene expression we treated neuronal and astroglial cultures with glucocorticoid receptor agonist (dexamethasone), mineralocorticoid (aldosterone), progesterone (progesterone), androgen (5α-dihydrotestosterone) and estrogen 2 (β-estradiol). The Dynamics of the Vaginal Microbiome During Infertility Therapy with In Vitro Fertilization-Embryo Transfer

Project description:Clinical treatment protocols for infertility with in vitro fertilization-embryo transfer (IVF-ET) provide a unique opportunity to assess the human vaginal microbiome in defined hormonal milieu. Herein, we have investigated the association of circulating ovarian-derived estradiol (E2) and progesterone (P4) concentrations to the vaginal microbiome. Thirty IVF-ET patients were enrolled in this study, after informed consent. Blood was drawn at four time points during the IVF-ET procedure. In addition, if a pregnancy resulted, blood was drawn at 4-to-6 weeks of gestation. The serum concentrations of E2 and P4 were measured. Vaginal swabs were obtained in different hormonal milieu. Two independent genome-based technologies (and the second assayed in two different ways) were employed to identify the vaginal microbes. The vaginal microbiome underwent a transition with a decrease in E2 (and/or a decrease in P4). Novel bacteria were found in the vagina of 33% of the women undergoing IVF-ET. Our approach has enabled the discovery of novel, previously unidentified bacterial species in the human vagina in different hormonal milieu. While the relationship of hormone concentration and vaginal microbes was found to be complex, the data support a shift in the microbiome of the human vagina during IVF-ET therapy using standard protocols. The data also set the foundation for further studies examining correlations between IVF-ET outcome and the vaginal microbiome within a larger study population.

Low plasma levels of miR-451a, miR-223-3p, and miR-27a-3p predict adverse prognosis in myelodysplastic syndromes

Project description:We investigated the spectra of circulating miRNAs in plasma of myelodysplastic syndromes (MDS) patients. Peripheral blood plasma from MDS patients with different risk scores was used for Agilent miRNA expression microarray analysis to define miRNA profile and to find miRNAs with discriminatory levels for lower risk and higher risk MDS. Results were further validated using droplet digital PCR on a larger cohort, enabling absolute quantification of plasma miRNAs and defining miRNAs with prognostic value for the disease. We analyzed expression profile of circulating miRNAs in plasma from 21 individuals: 7 controls and 14 MDS patients.

RNA-Seq of human cell lines Ishikawa, MCF7 and T47D treated with estrogen, progesterone and their antagonists

Project description:In this study the effect of estrogen (E2) and progesterone (P4) and their antagonists Tamoxifen, and Mifepristone (RU486) were assesed in endometrial epithelial cell line Ishikawa and compared to breast cancer cell lines MCF7 and T47D. Concentrations used for E2 and P4 treatments were 10 -8 M and for antagonists 10 -6 M. RNA-Sequencing (Illumina GenomeAnalyzer IIe) was applied to study global gene expression in three different cell lines. Expression values after hormonal or antagonist treatments in two different time points (3h and 12h) were compared to non-treated cells.
