Metabolites in contents of small intestine in wild type and DAOG181R/G181R mice

ABSTRACT: To investigate if DAO regulates metabolites in intestinal lumen, we compared metabolites in contents of small intestine in wild type and DAOG181R/G181R mice. All mice have C57BL/6 background and 8 weeks of age.

A non-dividing population with high pyruvate dehydrogenase kinase activity drives metabolic heterogeneity and tumorigenesis in the intestine

Project description:Although reprogramming of cellular metabolism is a hallmark of cancer, little is known about how metabolic reprogramming contributes to early stages of transformation. Here, we show that the histone deacetylase SIRT6 regulates tumor initiation during intestinal cancer by controlling glucose metabolism. Loss of SIRT6 results in increased number of intestinal stem cells (ISCs), which translates into enhanced tumor initiating potential in APCmin mice. More importantly, we found a metabolic compartmentalization within the intestinal epithelium and adenomas, where a rare population of cells exhibit features of Warburg-like metabolism characterized by high pyruvate dehydrogenase kinase (PDK) activity. Our results show that these cells are quiescent cells expressing +4 ISCs and enteroendocrine markers. Active glycolysis in these cells suppresses ROS accumulation and enhances their stem cell and tumorigenic potential. Our studies reveal that aerobic glycolysis represents a highly heterogeneous feature of cancer, and more importantly, they indicate that this metabolic adaptation occurs in non-dividing cells, suggesting a role for the Warburg effect beyond biomass production in tumors.

Development of a primary human Small Intestine-on-a-Chip using biopsy-derived organoids

Project description:Here we describe a method for fabricating a primary human Small Intestine-on-a-Chip (Intestine Chip) containing epithelial cells isolated from healthy regions of intestinal biopsies. The primary epithelial cells are expanded as 3D organoids, dissociated, and cultured on a porous membrane within a microfluidic device with human intestinal microvascular endothelium cultured in a parallel microchannel under flow and cyclic deformation. In the Intestine Chip, the epithelium forms villi-like projections lined by polarized epithelial cells that undergo multi-lineage differentiation similar to that of intestinal organoids, however, these cells expose their apical surfaces to an open lumen and interface with endothelium. Transcriptomic analysis also indicates that the Intestine Chip more closely mimics whole human duodenum in vivo when compared to the duodenal organoids used to create the chips. Because fluids flowing through the lumen of the Intestine Chip can be collected continuously, sequential analysis of fluid samples can be used to quantify nutrient digestion, mucus secretion and establishment of intestinal barrier function over a period of multiple days in vitro. The Intestine Chip therefore may be useful as a research tool for applications where normal intestinal function is crucial, including studies of metabolism, nutrition, infection, and drug pharmacokinetics, as well as personalized medicine.

Project description:Histone dimethyl transferase WHSC1 drives the transcription of MHC-I machinery in mouse and human colorectal cancer cells (CRCs); thus, WHSC1 downregulation potentiates CRCs to escape from cytotoxic CD8+ T cell responses. WHSC1 directly interacts with MHC-I transactivator, NLRC5 to selectively stimulate MHC-I gene expression. Thus, silencing Whsc1 diminished MHC-I level, impaired anti-tumor immunity and blunted immunotherapy efficacy.

Global gene expression profile of small intestine from wild-type and NKCC1 (SLC12A2)-knockout mice.

Project description:The objective of this study was to identify changes in gene expression levels between wild-type and NKCC1-knockout small intestine. 6 wild-type and 6 NKCC1-knockout small intestine RNA samples were compared. Total RNA was collected from both male and female 8-week old wild-type and NKCC1-null mice on an inbred FVB\N background. All comparisons were done between wild-type and NHE4-null samples from age- and gender-matched mice.
