Project description:Purpose: RNAseq analyses were conducted to screen for the genes undergoing transcriptional changes either in the liver of high-fat-diet (HFD)-induced obese mice or in the liver of Lepr-deficient db/db mice compared to the livers of the respective control mice Methods: C57BL/6 wild-type male mice were fed on high-fat diet (HFD) or a low-fat diet (NCD) for 18 weeks starting from 6 weeks of age, and the livers were collected at 24 weeks of age at ad libitum-fed condition.Misty/misty or db/db were sacrificed at ad libitum-fed condition at 10weeks and the liver was collected. Results: 2079 genes and 1085 genes were identified in high-fat-diet fed mice and db/db mice, respectively.

Project description:We propose comparing liver gene expression of WT and female ERKO mice early in the high-fat feeding period to animals fed a regular chow diet. Analyzing liver tissue before the fatty liver disease phenotype becomes severe will allow identification of target genes which may be causal. Comparison of regular chow fed WT animals to high fat fed WT animals will allow for identification of hepatic genes up-regulated in response to high fat feeding. Comparison of regular chow fed WT animals to regular chow fed ERKO animals will help clarify hepatic gene expression patterns that may be implicated in increased susceptibility to weight gain and glucose intolerance. Comparison of high fat fed WT animals to high fat fed ERKO animals will provide insight into genes that could be implicated in leading to increased fat accumulation in the liver over time during high fat feeding. Finally, comparison of regular chow fed ERKO animals to high fat fed ERKO animals will help identify genes that may be contributing to increased liver fat accumulation in response to high fat feeding in these animals.

Effect of rimonabant on gene expression of visceral and subcutaneous adipose tissues: high fat diet

Project description:Mice were fed for 6 months with a normal chow (NC) or a high fat diet (HFD). After 6 months of diet, high fat fed mice with Low, Medium and High body weight, but with similar glucose intolerance, were selected. These selected mice as well as NC mice were then treated with Rimonabant or Vehicle for 1 month. After treatment, mice were sacrificed and visceral and subcutaneous adipose tissues were collected and immediately frozen in liquid nitrogen. Total RNA from each sample was further extracted, purified, quality-controled before and after amplification, Cy5-labeled and co-hybridized with a Cy3-labeled mouse Universal Reference total RNA on the mouse 17K microarray. Project description:We report the results from RNA-seq analysis of perivascular adipose tissues (PVAT) collected from mesenteric arteries from Dahl S rats. Animals were on a control or a high fat diet for 24 weeks. At the moment of collection, rats fed high fat diet were hypertensive. In contrast, rats on the control diet were normotensive.
