Project description:We performed scRNA-seq analysis on the livers from the mice treated with a pan-integrin αV inhibitor, CWHM-12, after partial inferior vena cava ligation (pIVCL), a mouse model of congestive hepatopathy. The scRNA-seq analysis showed that integrin αV inhibition decreased the expression of YAP/TAZ target genes in pericentral LSECs and collagen expression in HSCs, caused by hepatic congestion.
Project description:Utilizing the 10X Genomics Cell Ranger (Pipeline Version 3.1.0) platform, this study investigates the impact of osteocyte ablation on skin aging. Skin and bone share significant structural and molecular homology, including compositional architecture, cellular origins, and signaling pathways. Both tissues exhibit parallel degenerative changes during natural aging and under pathological conditions, characterized by collagen depletion leading to osteoporosis and skin atrophy. Through scRNA-seq analysis of skin following osteocyte knockout, our research provides novel insights into the regulatory role of osteocytes in skin aging.
Project description:Langerhans cells (LC) in skin help initiate the immune response to locally presented antigens. We performed high-resolution single-cell RNA-sequencing (scRNAseq) analysis for antigen presenting cells including LC in normal mouse skin, and in mouse skin expressing the human papillomavirus (HPV) 16 E7 oncogene. Ear skin was collected from normal and trangenic mice. Dissociated CD45+ cells were processed for scRNA-seq using the 10X Genomics Chromium 3' gene expression kit (v2).
