Project description:The Streptophyta include unicellular and multicellular charophyte green algae and land plants. Colonization of the terrestrial habitat by land plants was a major evolutionary event that has transformed our planet. So far lack of genome information on unicellular charophyte algae hinders our understanding of the origin and the evolution from unicellular to multicellular life in Streptophyta. This work reports the high-quality reference genome and transcriptome of Mesostigma viride, a single-celled charophyte alga with a position at the base of Streptophyta. There are abundant segmental duplications and transposable elements in M. viride, which contribute to a relatively large genome with high gene content compared to other algae and early diverging land plants. This work identifies the origin of genetic tools that multicellular Streptophyta have inherited and key genetic innovations required for evolution of land plants from unicellular aquatic ancestors. The findings shed light on the age-old questions of the evolution of multicellularity and the origin of land plants.

Project description:Alternation of generations, in which the haploid and diploid stages of the life cycle are each represented by multicellular forms that differ in their morphology, is a defining feature of the land plants (embryophytes). Anciently derived lineages of embryophytes grow predominately in the haploid gametophytic generation from shoot apical meristems (SAMs) that give rise to photosynthetic structures with either leaf-like organs or a ribbon-like thallus. More recently evolved plant lineages have multicellular SAMs, and photosynthetic shoot development is restricted to the sporophyte generation. The molecular genetic basis for this evolutionary shift from gametophyte dominant to sporophyte dominant life cycles remains a major question in the study of land plant evolution. We used laser microdissection and next generation RNA sequencing to address whether angiosperm meristem patterning genes expressed in the sporophytic SAM of Zea mays are expressed in the gametophytic SAMs, or in the non-meristematic sporophytes, of the model bryophytes Marchantia polymorpha and Physcomitrella patens.

Project description:Colonization of land from marine environments was a major transition for biological life on Earth, and intertidal adaptation was a key evolutionary event in the transition from marine- to land-based lifestyles. Multicellular intertidal red algae exhibit the earliest, systematic, and successful adaptation to intertidal environments, with Porphyra sensu lato (Bangiales, Rhodophyta) being a typical example. We used proteomic analyse to reveal the complex regulation of rapid responses to intertidal dehydration/rehydration cycling within Neoporphyra haitanensis. These adaptations include rapid regulation of its photosynthetic system, a readily available capacity to utilize ribosomal stores, an excess of methylation supply to rapidly synthesize proteins, and a strong anti-oxidation system to dissipate excess redox energy upon exposure to air. These novel insights into the unique adaptations of red algae to intertidal lifestyles inform our understanding of adaptations to intertidal ecosystems and the unique evolutionary steps required for intertidal colonization by biological life.

Project description:The Zygnematophyceae are the closest algal relatives of land plants and hence interesting to understand land plant evolution. Species of the genus Serritaenia have an aerophytic lifestyle and form colorful, mucilaginous capsules, which surround the cells and block harmful solar radiation. Under laboratory conditions the production of this “sunscreen mucilage” can be induced by ultraviolet B radiation. The present dataset reveals insights into the cellular reaction of this alga to UV radiation (a major stressor in terrestrial habitats) and allows for comparisons with other algae and land plants to draw evolutionary conclusions.
