Project description:Human aortic endothelial cells were stimulated by lysophosphatidylcholine (LPC) (10μM) with or without interleukin 35 (IL-35) (10ng/mL) or IL-10 (10ng/mL) for 18 hours. Total RNAs were extracted from samples, then mRNA and non-coding RNAs were enriched by removing rRNA from the total RNA. The library was sequenced by Illumina HiSeq4000 using PE100 strategy and the reads were mapped to the human hg19 reference genome.

Project description:Vascular Innate immune cells, such as macrophages, elicit long-term T cell-, and B cell- immune responses, by expressing danger-associated molecular pattern (DAMP) receptors, T cell co-stimulation receptors and presenting antigens to the adaptive system through major histocompatibility complex (MHC)-II. Here, we investigated whether human aortic endothelial cells (HAECs) could also be “transdifferentiated” into innate immune cells after treatment of hyperlipidemia-upregulated DAMP molecules, lysophosphatidylinositol (LPI). HAECs were treated with vehicle control or lysophosphatidylinositol (16:0) (10µM) for 18 hours.

Project description:TNFα is a potent cytokine to mediate inflammatory response by activation of the master transcription factor NF-kB. Endothelial cells are important participants in inflammatory responses in animals. NF-kB is a major mediator to activate endothelial cells by inducing multiple pro-inflammatory genes in response to TNFα. The process of transcriptional activation is dependent on the context of chromatin interactions/loops. However, the chromatin conformation in response to the cytokine challenge TNFα in human aortic endothelial cells has not been described. Our approach characterized chromatin interactions from active enhancers to promoters as well as active promoters to promoters on a genome-wide scale and highlighted important enhancer candidates for target genes.

Lysophosphatidylcholine acyltransferase 1 controls the mitochondrial reactive oxygen species generation and survival of the retinal photoreceptor cells [P8 photoreceptor]

Project description:Due to the high energy demands and characteristic morphology, retinal photoreceptor cells require a specialized lipid metabolism for survival and function. This study focused on the roles of saturated fatty acids and their metabolism. A frameshift mutation of lysophosphatidylcholine acyltransferase 1 (Lpcat1), introducing saturated fatty acids into lysophosphatidylcholine to produce disaturated phosphatidylcholine (PC), has been reported to cause spontaneous retinal degeneration in mice (rd11 mice).　In this study, we performed a detailed characterization of Lpcat1 in the retina and found that Lpcat1 deficiency induces light-independent and photoreceptor-specific apoptosis in mice. Lipidomic analyses of the retina and isolated photoreceptor outer segment (OS) suggested that loss of Lpcat1 affects disaturated PC production and the proper cellular fatty acid flux, presumably by altering saturated fatty acyl-CoA availabilities. Furthermore, we demonstrated that Lpcat1 deletion increased mitochondrial reactive oxygen species (ROS) levels in photoreceptor cells, but not in other retinal cells without affecting the OS structure and trafficking of OS-localized proteins. These results suggest that the LPCAT1-dependent production of disaturated PC is critical for metabolic adaptation during photoreceptor maturation. Our findings highlight the therapeutic potential of saturated fatty acid metabolism in photoreceptor cell degeneration-related retinal diseases.
