Project description:Lactating cows experience transient metabolic stresses during postpartum which results in abnormal concentrations of non-esterified fatty acids and beta-hydroxybutyrate that can affect embryo-maternal communication and ultimately reproductive success. We hypothesize that metabolic challenges in lactating cows influence DNA methylation changes of the embryos prior to implantation affecting genes involved in embryo developmental competency. Therefore, we compared whole genome bisulfite sequencing (wgbs) of morulas derived from in vitro produced 2-4 cell embryos and transferred in the oviduct of metabolically profiled lactating cows until day 7. Similar stage of in vitro production (IVP) embryos were transferred to the oviduct of nulliparous heifers as a metabolically unchallenged control. Bisulfite-Seq DNA libraries were generated from groups of five morulas using an EZ DNA methylation-direct kit and Pico-Methyl seq library preparation kit (Zymo Research), and parallel sequenced using the illumine HiSeq2500 system at the institute of Genome Biology, Leibniz-Institute for Farm Animal Biology (FBN), Dummerstorf, Germany.

Project description:Lactating cows experience transient metabolic stresses during postpartum which results in abnormal concentrations of non-esterified fatty acids and beta-hydroxybutyrate that can impact oocyte maturation and ultimately reproductive success. We hypothesize that metabolic challenges in lactating cows influence DNA methylation changes of the oocyte affecting genes involved in oocyte developmental competency. Project description:In dairy cows, the transition from dry and pregnant to non-pregnant and lactating creates metabolic and immunological strain, which means many cows succumb to metabolic and infectious disease. This peripartum period, from three weeks pre-calving to three-weeks post-calving, is also known at the transition period. Metabolic health status during the transition period is determined by plasma concentrations of non-esterified fatty acids (NEFA), plasma β-hydroxybutyrate (BHBA), and liver triacylglycerol (TAG). High concentrations of these indicate cows that are under metabolic stress during the transition period and are, therefore, more likely to experience health conditions, also known as transition failure. Neutrophils, phagocytic cells of the innate immune response, are fundamental for fighting infectious agents.

Hepatic urea, creatinine and uric acid metabolism in dairy cows with divergent milk urea concentrations

Project description:Milk urea concentration is an indicator for dietary nitrogen (N)-supply and urinary nitrogen excretion. Dairy cows with high (HMU) compared to low milk urea (LMU) concentration have greater plasma urea, creatinine and uric acid concentrations, but if the liver metabolism accounts for these differences is unknown. Eighteen HMU and 18 LMU cows were fed either a diet with a low (LP) and normal (NP) crude protein concentration. An N-balance study was performed and a 13C-urea bolus was administered followed by a series of blood samplings. Liver samples were analysed by 2D-gel-based proteomics and real-time RT-PCR. Although HMU cows had a higher urea pool, plasma urea, uric acid, and hippuric acid concentrations, these differences were not associated with increased activation of the hepatic urea cycle or non-urea nitrogen metabolite pathways. Instead, HMU cows had a higher oxidative stress level. Conclusively, other factors than hepatic urea metabolism account for the milk urea concentration. Despite higher plasma urea concentrations on the LP diet, urea cycle mRNA expression was not affected, indicating it is not controlled at transcriptional level. Feeding the LP diet resulted in higher expressions of enzymes catabolizing fatty acids, which is likely due to diminished microbial growth and insufficient energy supply.

Project description:Homeorhetic mechanisms assist dairy cows to transition from pregnancy to lactation. In some cows this is less successful and they develop severe negative energy balance, placing them at risk of metabolic and infectious disease and reduced fertility. We placed Holstein Friesian cows into metabolically BALANCED or IMBALANCED clusters using as biomarkers measurements of plasma nonesterified fatty acids, β-hydroxybutyrate, glucose and IGF-I collected at 14 and 35 day in milk (DIM). To determine underlying mechanisms associated with their metabolic and immune dysfunction, we performed stranded total RNA sequencing using liver biopsy collected from some of the animals.
