Project description:We performed RNA seq to elucidate differential gene expression in A549 treated with Feiyanning (FYN). A549 cells were treated with FYN, then cultured for 24 h. Total RNA were extracted using TrizoI, and mRNA were retain using Oligotex mRNA Mini kit. The mRNA were fragmented into small fragments and reverse transcripted into cDNA. Next, the cDNA fragments were purified with Ampure beads, end repaired, poly(A) added, and ligated to Illumina sequencing adapters. Then User enzyme was used to digest the second-strand cDNA. The products were amplified, and sequenced.

Knockdown TIMM13 in A549 lung cancer cell line

Project description:We depleted TIMM13 using short interfering RNA (siRNA) in A549 lung cancer cells, and found that silencing of TIMM13 could significantly inhibit A549 cell proliferation. Thus, we conducted RNA-seq in the A549 cells transduced with either nontargeting siRNA or two distinct TIMM13-specific siRNAs. We observed widespread gene expression change in A549 cells upon TIMM13 knockdown.

MK886 treatment of H720 non-small cell lung cancer cell line

Project description:In this experiments different treatments were applied to lung cancer cell lines 1)H720 cell line suspension was treated by 1 micromole of 5-lipoxygenase activating protein inhibitor MK886 serum-free in TIS medium for 24 h. 2) A549 cell line was treated as H720 by MK886 3) A549 was treated by 100 nM of silencing cocktail (siRNA) prepared against the 4-th exon of the gene and generated by RNASEIII digestion of double stranded RNA. The cocktail was prepared by Ambion technology, see the manufacturer's protocol. The observed silencing was >90% 4) Based on Oligoengine vectors, silencing constructs were created against HRPA2B1 transcript(HNRPA2B1 is a subunit of spliceosome). Beta arrestin1 associated genomic regions in nicotine induced Non-Small Cell Lung Carcinoma (NSCLC) cell line A549

Project description:The multifunctional scaffolding protein M-NM-2-arrestin-1 plays a vital role in mediating the proliferative effects of nicotine through nAChR signaling. M-NM-2-arrestins were initially known as negative regulators of GPCR mediated signaling as they promote internalization and desensitization of GPCRs. However, new roles of M-NM-2-arrestins in receptor trafficking and signaling have been discovered in recent years. They are known to regulate signaling through a number of receptors such as Notch, endothelin A receptor, frizzled, smoothened and the nicotinic cholinergic receptors. Studies from our lab revealed that nAChR signaling induces the translocation of M-NM-2-arrestin-1 to the nucleus, in a Src dependent manner, where it directly binds to the proliferative E2Fs. Furthermore, the nuclear translocation of M-NM-2-arrestin-1 results in recruitment of p300 to E2F1 regulated proliferative promoters facilitating histone acetylation and transcription of these promoters. Given the role of M-NM-2-arrestin-1 in nicotine induced gene expression, we attempted to explore the global association of M-NM-2-arrestin-1 to the genomic regions upon nicotine stimulation by ChIP sequencing. It was found that M-NM-2-arrestin-1 is recruited on the promoters of many genes that regulate EMT as well as other regulatory pathways. In this assay, M-NM-2-arrestin-1 was found to be associated with the promoter regions of genes such as ZNF768, ZNF131, CSF3R, HMGA2, TAL1, RCC1, NKX2-4 etc in response to nicotine stimulation.
