Project description:Using a human-hamster radiation hybrid panel (G3), we have mapped loci regulating gene expression due to copy number increase. Copy number from radiation hybrid clones were measured relative to the A23 hamster recipient line. Expression from 79 radiation hybrid clones were measured in duplicate and compared to expression of the A23 hamster recipient line (GA_illumina_expression1 and GA_illumina_expression2). Expression of liver, kidney and heart from human and hamster are also included (GA_illumina_expression_R).

Project description:Plasma derived extracellular vesicles (EVs) have emerged as critical mediators of complications following traumatic injuries and after exposure to radiation. In response to injury, the cargo of these EVs is altered and can contain potent cargo that can influence cell function, including alterations in miRNA cargo. Here, we utilized NanoString's mouse miRNA panel to compare the alterations in miRNA content following exposure to 2 or 9 Gys of whole-body irradiation isolated 3 days after exposure and compared these changes to sham mice.

Project description:We have mapped regulatory loci for nearly all protein coding genes in the mammalian genome using microarray measurements from a panel of mouse/hamster radiation hybrids. Proof of principle transfection experiment. Pcdh7 (a gene identified as being a regulator of many genes) is transfected into A23 and HEK cells and compared to A23 and HEK cells transfected with an empty vector. Experiment Overall Design: Five replicas of RNA from the A23 cell line vs. itself. These arrays serve as a control to determine the power to detect cis ceQTLs in this experiment.
