Project description:FOXO1, a member of the FOXO forkhead type transcription factors, is markedly up-regulated in skeletal muscle during atrophy. Previously, we created transgenic mice specifically overexpressing FOXO1 in skeletal muscle (FOXO1 Tg mice). These mice weighed less than the wildtype control mice, had a reduced skeletal muscle mass. In this study, to better understand changes in skeletal muscle during atrophy, we performed a microarray analysis of skeletal muscle in wild-type control and FOXO1 Tg mice. The microarray data shows that in the skeletal muscles of FOXO1 Tg mice, gene expression of PGC-1β, a transcriptional regulator whose increased expression activates energy-expenditure-related genes in skeletal muscles, is decreased.
Project description:Analysis of skeletal muscle miRNA expression from type 2 diabetic volunteers before and after 16 weeks of chronic exercise training (two groups, one undergoing aerobic ecercise and the other resistance training exercise) A biopsy was collected from the right Vastus Lateralis under local anaesthesia and RNA was isolated from muscle (~10 mg) using mirVanaM-bM-^DM-" Kit , Low molecular weight RNA was biotin-labeled with FlashTagM-bM-^DM-" Biotin HSR RNA Labeling Kit. 21.5 M-BM-5l of high-quality biotin-labeled miRNA sample was hybridized to Affymetrix Gene-ChipM-BM-. miRNA 2.0, stained on Affymetrix Fluidics station 400 and scanned with a Hewlett Packard G2500A gene Array Scanner. AffymetrixM-BM-. miRNA QC Tool v 1.1.1.0 was used for the data summarization, normalization, and microarray quality control.
Project description:MKR mice is a Type 2 Diabetic mice, which was created by expressing mutation in IGF1 receptor in the skeletal muscle, and is widely used in diabetes research. Gene expression differences between MKR mice and Healthy (Wild type) mice are poorly understood. We used microarrays to detail the differences between gene expression in liver, fat, and skeletal muscle of MKR and Healthy mice. Male 10-weeks old FVB/N M-bM-^@M-^S MKR and wild type mice were used for the microarray studies. 3 biological replicates for each tissue type and for both MKR and WT mice were used in the studies.
