Project description:Saccharomyces cerevisiae has been used as a secretion host for production of various products, including pharmaceuticals. However, few antibody molecules have been functionally expressed in S. cerevisiae due to the incompatible surface glycosylation. Our laboratory previously isolated a group of yeast mutant strains with different α-amylase secretory capacities, and these evolved strains have showed advantages for production of some heterologous proteins. However, it is not known whether these secretory strains are generally suitable for pharmaceutical protein production. Here, three non-glycosylated antibody fragments with different configurations (Ran-Fab fragment Ranibizumab, Pex-the scFv peptide Pexelizumab, and Nan-a single V-type domain) were successfully expressed and secreted in three background strains with different secretory capacities, including HA (wild type), MA (evolved strain), and LA (evolved strain). However, the secretion of Ran and Nan were positively correlated with the strains’ secretory capacity, while Pex was most efficiently secreted in the parental strain. Therefore, transcriptional analysis was performed to explore the fundamental changes triggered by the expression of the different pharmaceutical proteins in these selected yeast strains.

Project description:Expression profiles of 110 the central metabolism-related enzymes were obtained by the selected reaction monitoring (SRM) assay methods using LC-MS/MS from the wild type (BY4742), a GCR2 gene deletion strain, and 29 single-gene deletion strains lacking enzyme genes responsible for central carbon metabolism (including CIT1, ENO1, FBP1, GCR2, GND1, GPD1, GPM2, HOR2, HXK1, HXK2, IDH1, IDH2, IDP1, LPD1, MAE1, MDH1, MDH2, PDA1, PDC1, PFK1, PYC2, RPE1, TAL1, TDH1, TDH2, TDH3, TKL1, TPS1, TPS2, and ZWF1 genes).
The central carbon metabolism is strictly controlled by modulation of enzyme expressions to maintain an essential system of living organisms. In this study, metabolic safety mechanisms in the model organism, Saccharomyces cerevisiae, were investigated by direct determination of enzyme expression levels. Targeted proteome analysis of 31 S. cerevisiae wild type and mutant strains revealed that at least 30% of the observed variations in enzyme expression levels could be explained by global regulatory mechanisms. Co-expression analysis revealed that expression levels of enzymes involved in trehalose metabolism and glycolysis changed in a coordinated manner under the control of the transcription factors for global regulation.

Project description:The homologous Ace2 and Swi5 transcription factors of Saccharomyces cerevisiae have identical DNA-binding domains, and both are cell cycle regulated. There are common target genes, as well as genes activated only by Ace2 and other genes activated only by Swi5. Keywords: genetic modification RNA was isolated from four strains: wild type, ace2 gene deletion, swi5 gene deletion, and the ace2 swi5 double gene deletion. RNAs from the three mutant strains were compared to wild type RNA in a microarray hybridization experiment.
