The effects of heat stress at 41M-BM-0C for 30 min (mild hyperthermia) on the gene expression in human cervical squamous cell carcinoma HeLa cells were investigated using an Affymetrix GeneChip system. Human cervical squamous cell carcinoma HeLa cells were treated with heat stress (41M-BM-0C for 30 min) and followed by incubation for 0, 1, or 3 h at 37M-BM-0C. Non-treated cells were served as control. Total RNA samples were prepared from the cells, and quality of the RNA was analyzed using a Bioanalyzer 2100. Gene expression was monitored by an Affymetrix GeneChipM-BM-. system with a Human Genome U133-plus 2.0 array. Sample preparation for array hybridization was carried out as described in the manufacturerM-bM-^@M-^Ys instructions.

Project description:Persistent infection by high-risk human papillomaviruses (HPVs) is associated with the development of cervical cancer and a subset of anogenital and head and neck squamous cell carcinomas. Abnormal expression of cellular microRNAs (miRNAs) plays an important role in the development of cancer, including HPV-related tumors. MiRNA expression profile was investigated by microrray analysis in the HPV-positive cervical cancer cell lines SiHa (HPV16-positive cell line derived from a cervical squamous cell carcinoma), CaSki (HPV16-positive cell line derived from a metastatic cervical epidermoid carcinoma), and HeLa (HPV18-positive cell line derived from a cervical adenocarcinoma) and compared with primary HFKs and C33a (HPV-negative cervical cell line).

The data suggested similar expression profiles to each other. Small RNA samples prepared from HeLa cells exposed to a mild hyperthermia (40M-BM-:C) as well as normal temperature (37M-BM-:C) were examined by means of GenopalM-BM-.-MICH07 DNA chips.
