Overall design: Mesophyll mRNA profiles of wild type (WT) Ammopiptanthus mongolicus leaf (normal,drought stress,cold stress) were generated by deep sequencing, in triplicate, using Illumina GAIIx.
Project description:Ammopiptanthus mongolicus is an ecologically important desert plant. To understand the role of miRNA genes in draught and cold stress adaptation, RNA-seq of small RNA in Ammopiptanthus mongolicus was performed. Overall design: To identify microRNAs and other small RNA in A. mongolicus, we generated five small RNA libraries using RNA isolated from drought stressed-roots (DT_R) and unstressed roots (CK_R), and RNA isolated from drought stressed-leaves (DT_L), cold stressed-leaves (CT_L) and unstressed leaves (CK_L). These small RNA libraries were then sequenced by the high throughput Illumina /Solexa sequencing platform.
Project description:Ammopiptanthus mongolicus is an ecologically important desert plant. miRNA are endogenous 20-22 nt small RNAs that play important roles in regulating gene expression in plants. To identify to the target genes of miRNA in Ammopiptanthus mongolicus, degradome sequencing was performed and a batch of transcripts were found to be targeted by miRNA. The result is helpful in understanding the roles on miRNA in plant development and environmental stress response. Overall design: A degradome sequencing library was constructed using pooled mRNA isolated from leaves and roots of A. mongolicus seedlings. The library was sequenced using Illumina Hiseq 2500 platform.
