Project description:Purpose: We here wanted to describe the gene regulation of Gigaspora rosea in association with phyllogenetically divergent plant hosts, and compare these results with gene regulation occuring in R. irregularis, the model arbuscular mycorrhizal fungus. Methods: mRNA from Medicago truncatula (legume), Brachypodium distachyon (grass) in association with G. rosea, and extra radical mycelium of G.rosea were sequenced by Illumina. Reads were mapped on a in-house de novo transcript assembly with the software CLC workbench. Fungal gene expression in the different host plants was compared to extra radical hyphae as reference. Results: Sets of 1891 and 1566 G. rosea genes were highly overexpressed (fold change >5 ; FDR <0,05 and experimental value difference > 10) , in M. truncatula and B. distachyon respectively compared to ERM, among which 802 of them were up-regulated in both plants. Non common up-regulated genes are mainly found non statistically robust in one condition. Conclusions: Our study represents the first transcriptomic analysis on several hosts for this fungal species. These results showed that the interaction between plants and AMF is highly conserved.

Reads were mapped on the genome assembly with the software CLC workbench. Fungal gene expression in the different plants was compared to extra radical hyphae as a control. Results: 529, 486 and 523 R. irregularis gene were highly overexpressed (fold change >5 ; FDR <0,05 and experimental value > l10l) in M. truncatula, B. distachyon and L. cruciata, respectively. Among those genes, 262 were induced in all hosts. qPCR validation on 32 genes supported these results in an extended set of hosts (Zea mays spp parviglumis, Pisum sativum, Marchantia paleacea).

Project description:Emergence of the symbiotic lifestyle fostered the immense diversity of all ecosystems on Earth, but symbiosis plays a particularly remarkable role in marine ecosystems. Photosynthetic dinoflagellate endosymbionts power reef ecosystems by transferring vital nutrients to their coral hosts. The mechanisms driving this symbiosis, specifically those which allow hosts to discriminate between beneficial symbionts and pathogens, are not well understood. Here, we uncover that host immune suppression is key for dinoflagellate endosymbionts to avoid elimination by the host using a comparative, model systems approach. Unexpectedly, we find that the clearance of non-symbiotic microalgae occurs by non-lytic expulsion (vomocytosis) and not intracellular digestion, the canonical mechanism used by professional immune cells to destroy foreign invaders. We provide evidence that suppression of TLR signalling by targeting the conserved MyD88 adapter protein has been co-opted for this endosymbiotic lifestyle, suggesting that this is an evolutionarily ancient mechanism exploited to facilitate symbiotic associations ranging from coral endosymbiosis to the microbiome of vertebrate guts.

Project description:Receptor-like kinases (RLKs) are key cell signalling components. The rice ARBUSCULAR RECEPTOR-LIKE KINASE 1 (OsARK1) functions during a stage post-arbuscule development in the arbuscular mycorrhizal (AM) association and belongs to an undefined subfamily of RLKs. Our phylogenetic analysis revealed that ARK1 has an ancient paralogue, ARK2. Single ark2 and ark1ark2 double mutants in rice showed a non-redundant AM symbiotic function for OsARK2. Global transcriptomics identified a set of genes co-regulated by the two RLKs suggesting OsARK1 and OsARK2 to orchestrate symbiosis in a common pathway. ARK lineage proteins harbour a newly identified SPARK domain in their extracellular regions which underwent parallel losses in ARK1 and ARK2 in monocots. This protein domain has ancient origins in the green lineage and defines additional overlooked groups of putative cell surface receptors

This shows that both proteins are functionally conserved in higher plants, suggesting an ancient function that was conserved during evolution. Here we show that NSP1 and NSP2 are indispensable for strigolactone biosynthesis in the legume Medicago truncatula as well as rice. Mutant nsp1-nsp2 plants hardly produce strigolactones. The lack of strigolactone biosynthesis coincides with strongly reduced DWARF27 expression in both species. Rice and Medicago represent distinct phylogenetic lineages that split ~150 million years ago. Therefore we conclude that regulation of strigolactone biosynthesis by NSP1 and NSP2 is an ancestral function conserved in higher plants. Since strigolactone biosynthesis is highly regulated by environmental conditions like phosphate starvation, NSP1 and NSP2 will be important tools in future studies on the molecular mechanisms by which environmental sensing is translated into regulation of strigolactone biosynthesis. As NSP1 and NSP2 are single copy genes in legumes, it implies that a single protein complex fulfills a dual regulatory function of different downstream targets; symbiotic and non-symbiotic, respectively.
