Project description:The goals of this study are to compare different gene expressions for Penicillium oxalicum wild type strain (WT), and Podot1 knockout strain (ΔPodot1) in different carbon sources. The deletion of Podot1 downregulated genes involved in the septin complex, extracellular region, and interspecies interaction between organisms when strains were cultivated with 2% glucose as carbon sources, and downregulated genes involved in cellulase activity, cellulose binding, glucosidase activity, and polysaccharide catabolic process when strains were cultivated with 1% microcrystalline cellulose and 1% wheat bran as carbon sources. We find the extracellular region was downregulated under both different carbon sources in ΔPodot1. This study provides the information that PoDot1 function are required in mycelial development and hydrolase activity of P. oxalicum.

Project description:The filamentous fungus Penicillium oxalicum can secret various enzymes for efficient saccharification of plant biomass materials. Expression of the constitutively active forms of transcriptional activators ClrB, XlnR and AraR could trigger the production of different sets of lignocellulolytic enzymes. Here, the transcriptomes of the three engineered strains were compared with that of wild type in the medium without carbon source.

The deletion of set2 upregulated genes involved in oxidation- reduction process, extracellular region, and plasma membrane ATP synthesis coupled proton transport both with 2% glucose or 1% cellulose and 1% wheat bran as carbon sources. We find the expression levels of 20 secondary metabolism gene clusters were upregulated or downregulated under different carbon sources in Δset2. This study provides the information that SET2 function are required in conidiation and hydrolase activity of P. oxalicum.

The deletion of laeA downregulated genes involved in oxidation- reduction process, alkaloid metabolic process, and transmembrane transport. We find the expression levels of seven secondary metabolism gene clusters (totally 28 clusters) were silenced inΔlaeA. This study provides the information that laeA function are required in conidiation and hydrolase activity of P. oxalicum. Examination of differential gene expressions by digital gene expression tag profiling in Penicillium oxalicum wild type strain and laeA knockout mutant strains in 24h and 60h in modified Czapek culture medium with 2% glucose as carbon resource. qRT–PCR validation was performed using SYBR Green assays.
Project description:The synthesis of cellulolytic enzymes by filamentous fungi are mainly regulated at the transcriptional level. The essential role of transcriptional activator ClrB/CLR-2 in the induction of cellulase genes has been reported in several fungal species. In this study, we identified the C-terminal region of ClrB in Penicillium oxalicum as a transcriptional activation domain using a yeast reporter gene system. Overexpression of a mutant ClrBID fusing the DNA-binding and transcriptional activation domains of ClrB together enabled cellulase production under cellulase non-inducing conditions. Strikingly, the ClrBID-overexpression strain produced cellulase in the presence of glucose at a level similar to that of reference strain on cellulose. Combination of ClrBID overexpression and deletion of carbon catabolite-repressor gene creA suggested an additive effect of the two manipulations on carbon catabolite de-repression. A similar modification of ClrB in Aspergillus niger also resulted in cellulase production on glucose. The results indicated that the post-transcriptional control of ClrB activity involves its central region, and provided a potential strategy for engineering Penicillium and Aspergillus strains for cellulase production on easy-to-use carbon sources.

Project description:The goals of this study are to compare different gene expressions for Penicillium oxalicum wild type strain (WT), Hat1 deletion strain (ΔHat1) and AmyR deletion strain (ΔAmyR) in 2% starch as carbon sources. The correlation analysis results between the various samples showed that the gene expression levels of the wild strain and the ΔHat1 strain were significantly different, and the gene expression levels between ΔAmyR and the wild strain were also significantly different. The deletion of Hat1 significantly up-regulates the expression levels of amylase genes of Penicillium oxalicum, and the absence of AmyR can significantly down-regulate the expression level of amylase genes genes, indicating that Hat1 and AmyR can cause a difference in amylase synthesis by affecting the expression of amylase genes. The information provided by this study indicates that Hat1 and AmyR functions are necessary for the amylase activity of Penicillium oxalicum.
