Project description:We report that there are differences in V gene usage in memory B cells and long-lived plasma cells after oral immunization. The hapten NP was conjugated to cholera toxin (CT) to create NP-CT, an antigen that is highly immunogenic after oral immunization. The NP hapten induces immune responses dominated by the 1-72 (VH186.2) heavy chain V region. The relationship between NP binding IgA antibody genes from memory B cells from spleen, MLN and Peyer´s patches and long-lived plasma cells from lamina propria and bone marrow that persisted 6-12 months after an oral immunization in three C57BL/6 mice with NP-CT was investigated. Extensive clonal overlap im clones was observed between long-lived memory cells from lamina propria and bone marrow, but very limited overlap was found between memory cells and plasma cells. The data suggest that memory and plasma cells formed through temporarily or anatomically separate processes.

Project description:To obtain insight into the genetic basis of the increase of functional activity of memory B cells over time, we compared the gene expression profiles of day 7 and day 40 NP-specific/IgG1 memory B cells, GC B cells and plasma cells in immunized WT mice and naïve B cells, before and after activation in vitro. Total RNAs in three replicates of NP-specific/IgG1 memory B cells, GC B cells and plasma cells in immunized WT mice at day 7 and 40 postimmunization and naïve follicular and marginal zone B cells, before and after activation in vitro, were amplified and labeled according to the Affymetrix's instructions. cRNA was hybridized to Mouse Genome 430 2.0 GeneChips (Affymetrix).

Project description:Little is known about the earliest transcriptomic events in plasma cell differentiation. Indeed, activation of UPR-affiliated gene expression is seen in in-vitro plasma cell differentiation prior to Blimp1 upregulation and in the absence of Xbp1 as we show in linked companion series. In this study, Blimp1 reporter mice were immunized with the T-independent antigen NP-LPS four days prior to spleen harvest and naïve B cells, NP-binding activated B cells and NP-binding plasma cells were analyzed by bulk RNA-seq. Here we report the activation of ER remodeling gene expression in antigen-specific activated B cells in-vivo prior to upregulation of Blimp1 transcription.

Project description:Next-generation sequencing (NGS) of antibody variable regions has emerged as a powerful tool in systems immunology by providing quantitative molecular information on polyclonal humoral immune responses. Reproducible and robust information on antibody repertoires is valuable for basic and applied immunology studies, thus it is essential to establish the reliability of antibody NGS data. Therefore, two conditions of immunological relevance were assessed for NGS reproducibility. First, we pooled spleen plasmablasts and plasma cells (CDR138-enriched) and bone marrow plasma cells (CD45R-depleted and CD138-enriched 4) of 1 mouse (1M) immunized with NP-CGG (chicken gamma globulin (CGG) conjugated to 4-hydroxy-3-nitrophenylacetyl) and sacrificed 14 days post-immunization (dpi). The thus created cell pool contained approximately 3 x 106 viable ASCs. Second, in order to model extreme antibody diversity, we repeated the same cell isolation procedure from 9 immunized mice (9M) resulting in an ASC pool of approximately 2.5 x 10^7 viable cells. From isolated cells, we recovered total RNA and used RT-PCR to amplify expressed rearranged IgG variable heavy VDJ genes; PCR was performed using a well-characterized and utilized primer set based on variable framework 1 region forward primers and one IgG constant region 1 reverse primer (covering all IgG isotypes) 43. Similarly to previously published methods (Vollmers et al., 2013, PNAS), Illumina adaptors were added during the PCR step by using a direct addition approach where adaptors are added at the 5’ end of the gene-specific portion of the primer set, thus circumventing the need for ligation of adaptors following PCR. For each of the two conditions (1M/9M), triplicates were prepared, where a triplicate signifies three separately indexed samples prepared from the same starting cDNA pool; thus variable region PCR was independently performed in each of the triplicates. All six samples (triplicates of 1M and 9M) were sequenced using the Illumina MiSeq platform with 250bp paired-end reads.

Project description:Selection of B cells subjected to hypermutation in germinal centres (GC) during T-dependent (TD) antibody responses yields memory cells and long-lived plasma cells that produce high affinity antibodies biased to foreign antigens rather than self-antigens. GC also form in T-independent (TI) responses to polysaccharide antigens but failed selection results in GC involution and memory cells are not generated. To date there are no markers that allow phenotypic distinction of T-dependent and T-independent germinal centre B cells. We have now compared the global gene expression of GC B cells purified from mice immunized with either TD or TI antigens and identified eighty genes that are differentially expressed in TD GC. Experiment Overall Design: QMxB6 F1 mice were immunised with T-independent antigens (NP-Ficoll) or T-dependent antigens (NP-CGG). T-dependent responses were analysed in carrier primed mice and non-carrier primed mice. The kinetics in carrier primed mice are comparable to those of TI-2 antigens, and thus, both of these groups were analysed 4 days after immunization with soluble antigen. The response in non-carrier primed mice peaks later, and was therefore analysed on day 10 after immunization. Germinal centre B cells were MACS enriched and then FACS sorted according to GL-7 and B220 expression. Sorted B cells from the relevant mice were pooled (4-10 mice per sample). Each sample thus contains B cells from different mice, and the replicates are biological, not technical replicates.
