Project description:Higher aridity and more extreme rainfall events in drylands are predicted under climate change. Yet it is unclear how changing precipitation regimes may affect nitrogen (N) cycling, especially in areas with extremely high aridity. Here we investigated soil N isotopic values (M-NM-415N) along a 3200 km aridity gradient and show a hump-shaped relationship between soil M-NM-415N and aridity index (AI) with a threshold at AI=0.32. Also, using a micro-array metageomics tool named GeoChip 5.0, we showed that Variations of nitrification and denitrification gene abundance along the gradient which provide further evidence for the existence of this threshold. Data support the hypothesis that the increase of gaseous N losses is higher than the increase of net plant N accumulation with increasing AI below AI=0.32, while the opposite is favoured above this threshold. Our results suggest the importance of N-cycling microbes in extremely dry areas and the different controlling factors of N cycling on the either side of the threshold.

Project description:We used microarrays to discern patterns of gene expression in response to global climate change factors on leaf tissue of an annual dicot, Geranium dissectum, growing in a natural grassland. Keywords: multifactorial global change treatments There are 5-8 biological replicates for each of the sixteen treatments - a total of 100 samples. The four global change factors at two levels are ambient or elevated CO2, ambient temperature or ambient + 1 degree C warming, natural rainfall (601.2 mm/yr) or natural rainfall supplemented with 50% of every storm added immediately following each storm with a 3 week season extension, ambient growth nitrogen or ambient supplemented with 7 g N m-2 y-1, supplied as Ca(NO3)2. There were 16 treatments applied in a nested design in the field.We hybridized the Geranium samples using cross-species hybridization to Arabidopsis cDNA arrays which had an Arabisopsis genomic DNA common reference in the Cy3 channel.

The climate variability hypothesis predicts that temperate ectotherms will be less vulnerable to climate warming than tropical ectotherms, due to their superior acclimatization capacity.We investigate thermal acclimation of three species of Takydromus lizards distributed along a broad latitudinal gradient in China, by studying metabolic modifications at the level of the whole organism,organ, mitochondria, metabolome, and proteome.

The climate variability hypothesis predicts that temperate ectotherms will be less vulnerable to climate warming than tropical ectotherms, due to their superior acclimatization capacity. However, metabolic acclimatization occurs over multiple levels of the biological hierarchy, from the enzyme and cellular level, through organ systems, to whole-organism metabolic rate. Previous studies have focused on one or a few biological hierarchy levels, leaving us without a general understanding of how metabolic acclimatization might differ between tropical and temperate species. Here, we investigate thermal acclimatization of 3 species of <em>Takydromus</em> lizards distributed along a broad latitudinal gradient in China, by studying metabolic modifications at the level of the whole organism, organ, mitochondria, metabolome and proteome. As predicted by the climate variability hypothesis, the 2 temperate species <em>T. septentrionalis</em> and <em>T. wolteri</em> had an enhanced acclimation response at the whole organism level compared to the tropical species <em>T. sexlineatus</em>, as measured by respiratory gas exchange rates. However, the mechanisms by which whole organism performance was modified was strikingly different in the 2 temperate species: widespread <em>T. septentrionalis</em> modified organ sizes, while the narrowly distributed <em>T. wolteri</em> relied on mitochondrial, proteomic and metabolomic regulation. We suggest that these 2 mechanisms of thermal acclimatization may represent general strategies used by ectotherms, with distinct ecological costs and benefits. Lacking either of these mechanisms of thermal acclimatization capacity, the tropical species is likely to have increased vulnerability to climate change.</p>
