Project description:Enterohaemorrhagic E. coli (EHEC) is a significant human pathogens that cause outbreaks of haemorrhagic colitis and haemolytic uremic syndrome. During infection, pathogens compete for iron with the host, and one mechanism by which EHEC obtains iron is through haem uptake and utilitisation which is encoded by the chu operon. We have demonstrated that the haem receptor chuA is regulated by the Crp-cAMP-dependent sRNA CyaR. We further demonstrate that activation of chuA by CyaR is independent of the chuA RNA-thermometer and termination by Rho. These results highlight the ability of regulatory sRNAs to integrate multiple environmental signals into a layered hierarchy of signal input.
Project description:We performed tRNAome sequencing to assess the tRNA changes of E.coli under oxidative stress. We found that the global translation inhibition is caused by global down-regulation of almost all tRNA species under oxidative stress. The translation elongation speed is resumed after the cells are fully adapted to the oxidative environment.

A four chip study using total RNA recovered from two separate cultures of Escherichia coli MG1655 K-12 M-bM-^HM-^Fhns/M-bM-^HM-^FstpA mutant strain under aerobic and anaerobic growth conditions. Each chip measures the expression level of 4,661 genes from Escherichia coli MG1655 K-12 using a high-density tiling array consisting of ~385,000 60mer probes spaced every 12 bp.
