Project description:The Chinese giant salamander (Andrias davidianus) is one of the most important ecological breeding species with distinct characteristics and is cultured in many locations throughout China. In the present study, the transcriptome of A. davidianus spleen tissue, that had challenged with Citrobacter freundii, was analyzed using Illumina sequencing technology. The result was compared to a heathy control group. After assembly and annotation, 128,540 transcripts were generated with a median length of 349 bp. Comparative expression analysis indicated 1,995 differentially expressed genes (DEGs), 812 of which were up-regulated and 1,183 were down-regulated. Furthermore, DEGs were classified into three gene ontology categories, 535 of which were annotated to 237 KEGG pathways. Finally, six immune-related DEGs involved in the immune-related pathways were randomly selected for scrutinization. This work provides valuable data for an improved understanding the defense mechanisms of A. davidianus against bacterial pathogens at the transcriptional level.
Project description:Chansu, which is prepared from the skin secretions of toad (Bufo bufo gargarizans Cantor), is widely used in traditional Chinese medicine (TCM). Being the principal bioactive constituents of ChanSu, bufalin (BFL) and cinobufagin (CBF) have been shown to possess anticancer properties. TCM confer bioactivities through the synergistic effect between potential active ingredients, so as to interfere with the development of the disease, and ultimately achieve the therapeutic effect. We found that the anticancer effect was significantly potentiated by co-treatment of BFL and CBF as compared to mono-treatment, suggesting their synergistic interaction. To reveal their synergistic mechanisms, metabolomic and lipidomic profiling based on liquid chromatography-mass spectrometry (LC���������MS) were utilized to delineate the responses in HepG2 cells after treatment with BFL and CBF individually or in combination. Metabolic pathways including methionine metabolism, energy metabolism, lipid metabolism and amino acid metabolism were modulated and subsequently lead to apoptosis and cell cycle arrest of HepG2 cells. In particular, the discrepant regulation of methionine metabolism between mono-treatment and co-treatment of BFL and CBF may account for their synergistic effect. Our study provided novel insights into the mechanistic links between cellular metabolism and synergistic effect, which may ultimately lead to better treatments for hepatoma.
Project description:Tadpoles of the anuran species Rana pirica can undergo predator-specific morphological responses. Exposure to a predation threat by larvae of the salamander Hynobius retardatus results in formation of a bulgy body (bulgy morph) with a higher tail. The tadpoles revert to a normal phenotype upon removal of the larval salamander threat. The objective of the present study was to use our own fabricated tadpole Rana pirica cDNA microarray to profile gene expression patterns during the predation threat.
