Project description:This paper describe the whole genome-wide analysis of differences in postnatal intestinal from 0 day to 21 days of piglets. Using Genome Arrayï¼We generated transcriptase profiles of intestine from the Landrace piglets at 0, 3, 8, 14ï¼21 days post natal (dpn). We identified 4014 differentially expressed transcripts during intestinal developing in Landrace piglets. Respectively, the Gene Ontology processes and the time sequence profile analysis of differentially expressed genes were constructed. In the course of growth , the mostly significant enriched GO category of profile mainly focus on folic acid transport, lipopolysaccharide-mediated signaling pathway, Momolybdopterin cofactor biosynthetic process,The down-regulation genes show the state of malnutritionplay at early growth in pig life. Project description:The study investigated the impact of environment on the composition of the gut microbiota and mucosal immune development and function at gut surfaces in early and adult life. Piglets of similar genotype were reared in indoor and outdoor environments and in an experimental isolator facility. Mucosa-adherent microbial diversity in the pig ileum was characterized by sequence analysis of 16S rRNA gene libraries. Host-specific gene responses in gut ileal tissues to differences in microbial composition were investigated using Affymetrix microarray technology and Real-time PCR. Experiment Overall Design: Animals were reared on the sow at an outdoor or indoor facility. Additional piglets from the indoor facility were transferred to individual isolator units at 24 hours of age, and given a daily dose of antibiotic cocktail for the duration of the study. Piglets were weaned at day 28. From day 29 onwards, piglets were fed creep feed ad libitum. Ileal tissue samples were excised from N=6 piglets per group at day 5, 28 and 56.

Project description:Porcine epidemic diarrhea virus (PEDV) has reemerged as the main pathogen of piglets due to its high mutation feature. Monolaurin (ML) is a natural compound with a wide range of antibacterial and antiviral activities. However, the role of ML in PEDV infection is still unknown. This study aimed to evaluate the effect of ML on the growth performance, intestinal function, virus replication and cytokine response in piglets infected with PEDV, and to reveal the mechanism through proteomics analysis. Piglets were orally administrated with ML at a dose of 100 mg/kg·BW for 7 days before PEDV infection. Results showed that although there was no significant effect on the growth performance of piglets, ML administration alleviated the diarrhea caused by PEDV infection. ML administration promoted the recovery of intestinal villi, thereby improving intestinal function. Meanwhile, PEDV replication was significantly inhibited, and PEDV-induced expression of IL-6 and IL-8 were decreased with ML administration. Proteomics analyses showed that 38 proteins were differentially expressed between PEDV and ML+PEDV groups, and were significantly enriched in the interferon-related pathways. This suggests ML could promote the restoration of homeostasis by regulating the interferon pathway. Overall, the present study demonstrated ML could confer a protective effect against PEDV infection in piglets, and may be developed as a drug or feed additive to prevent and control PEDV disease.

Project description:We demonstrated that a maternal antibiotic treatment can change intestinal development of the offspring piglets permanently by showing that maternal gestational antibiotic treatment affects intestinal development in offspring piglets for a period of at least seven weeks after the antibiotic treatment in the sows was finished. It was shown that immediately after birth the piglets from amoxicillin treated sows, showed upregulation of genes involved in processes related to ‘tight junctions’ and ‘immunoglobulins’. In addition, these piglets had significantly lower number of goblet cells. Together, this may lead to a gut wall that is more rapidly closed in piglets from amoxicillin treated sows, affecting the uptake of immunoglobulins and the intestinal development. Later in life, around weaning, gene expression and morphological data indicate that the crypts of piglets from amoxicillin treated sows deepen around weaning as an effect of the amoxicillin treatment which in combination with the upregulation of genes involved in cell cycle processes, ribosomal activity and protein degradation might imply that the intestinal development, the subsequent differentiation of cells or the timing of these processes was delayed by the maternal antibiotic treatment.

Comparisons between control and PCV2-inoculated pigs were done at five different time points: 1, 2, 5, 8, and 29 days post-inoculation. Experiment Overall Design: Seven-day-old caesarean-derived, colostrum-deprived piglets were distributed into two groups: control (n=8) and inoculated with 105.2 TCID50 of the Burgos PCV2 isolate (n=16). One control and 3 inoculated pigs were necropsied on days 1, 2, 5, and 8 post-infection (p.i.), the remaining pigs (4 of each group) were necropsied on day 29 p.i.

Project description:A gene expression topology of the developing, postnatal and diseased heart resulted in re-interpretation of ventricular remodeling in terms of rearrangement of key gene regulatory networks that are imbalanced, attenuated, or abnormally activated in the failing myocardium. The underlying principle, “Shaping the heart in development and disease” becomes a focus of current cardiovascular research with the goal of developing innovative diagnostic and therapeutic strategies to combat cardiovascular disease. In this line, our interests focus on regulatory molecular pathways and identification of novel candidate genes associated with ventricular remodeling in normal and pathological states. The general goal of the project was to establish a piglet model of heart failure induced by the cardiotoxic agent Doxorubicin. Transcriptomic analysis on Affymetrix GeneChip Porcine Genome Array was used to used to identified the genes associated with pathological ventricular remodeling. Six 8-day old piglets were randomized in two groups, the experimental group (n=3) received a single intravenous administration of 2.0 mg/kg Doxorubicin, whereas the control group (n=3) was injected with PBS. Piglets were euthanized 3 weeks later to harvest cardiac tissue for RNA isolation. Piglets were used in accordance with protocols approved by the Institutional Animal Care and Use Ethical Committee.
