Project description:Japonica rice cultivar Nipponbare was inoculated with wheat leaf rust (Puccinia triticina f. sp. tritici, non-host pathogen to rice) to compare gene expression profiles with mock-inoculated controls. Although eventually failed in invasion, leaf rust induced a set of rice genes that were distinctally up-regulated, some of those were confirmed by quantitative real-time PCR assays.

The proteome of germlings from urediniospores of Puccinia triticina

Project description:Wheat leaf rust is a serious fungal disease of wheat that causes annual losses and necessitates using fungicides for effective disease management. It is caused by Puccinia triticina which spreads by means of airborne urediniospores. When these germinate on the leaf surface, they form germ-tubes which enter the leaf through open stomates. Spores and germ-tubes represent the first fungal structures that the host can perceive during a rust infection. They therefore contain proteins that could potentially trigger early host defense responses. Using 2-DE to separate this proteome, we produced gels containing 173 spots in the pI range of 4-7 and identified 123 proteins. These were predominantly proteins involved in metabolic and cellular processes, but with a large number (77%) of novel proteins that could not be identified through homology matching Twenty four of these showed no homology to wheat sequences, making them good candidate PAMPs.

Project description:In this experiment the transcriptome reprogramming in wheat during host and nonhost interaction with Puccinia sp. was analyzed in a time-series approach. Ten days old wheat plants of cv. Renan were mock-inoculated or inoculated with P. triticina (Pt), BRW96258 isolate, or P. hordei (Ph), 1.2.1 isolate. After 12, 24, 36 and 48 hours first leaves were sampled. Total RNA was extracted using the RNeasy Plant Mini Kit (Qiagen, Hilden, Germany), the Ambion TURBO DNA-free DNase Kit was used for DNA elimination, and RNA was hybridized to Agilent 44k oligonucleotide arrays.

Transcript profiling of Lr1- and Lr34-mediated leaf rust resistance in wheat

Project description:The wheat gene Lr34 confers partial resistance to all races of Puccinia triticina, the causal agent of wheat leaf rust. However, the biological basis for the exceptional durability of Lr34 is unclear. The Affymetrix wheat genome array was used to identify wheat genes differentially expressed in a compatible interaction (Tc), an R-gene mediated incompatible interaction (Tc-Lr1), and a race non-specific resistance interaction (Tc-Lr34) in response to infection challenge by P. triticina race 1 at anthesis. Transcriptome interrogation was conducted by comparing mock- and P. triticina-inoculated leaves harvested at 3 and 7 days post inoculation (dpi). Keywords: Time course
