Project description:Toll-like receptors (TLRs) are key regulators of innate immune responses, and their dysregulation is observed in numerous inflammation-associated malignancies, including gastric cancer (GC). However, the identity of specific TLRs and their molecular targets which promote the pathogenesis of human GC is ill-defined. Here, we sought to determine the clinical utility of TLR2 in human GC. TLR2 mRNA and protein expression levels were elevated in 50% of GC patient tumors across multiple ethnicities. TLR2 was also widely expressed among human GC cell lines, and DNA microarray-based expression profiling was conducted on RNA from NUGC4 and AZ521 cells either non-stimulated or stimulated with a combination of 10g/ml Pam3Cys-Ser-(Lys)4 and FSL-1. Only NUGC4 and AZ521 were analyzed for publication.

Project description:Monocytes and monocyte derived macrophages, besides natural killer (NK) cells, are crucial for elimination of senescent cells. There is evidence that TLR2/6 together with the lipid receptor CD36 may be the pattern recognition receptor that are most involved in the sensing of epitopes connected to cellular aging, senescence and severe cellular distress. Additionally, the interplay of these scavenger receptors is required for uptake of TLR2/6 agonist FSL-1. Therefore, we investigated whether lysophospholipids would induce or interfere with (TLR2/6 mediated) gene regulation in U937 monocytes. Cells were or were not pre-treated with lysophospholipids (10µM) for 1 hour and then stimulated with the TLR2/6 ligand (1µg/ml) for 2 hours.

Project description:In order to elucidate transcriptional and metabolic networks associated with Lys metabolism, we utilized developing seeds as a system in which Lys synthesis could be stimulated developmentally without application of chemicals and coupled this to a T-DNA insertion knockout mutation impaired in Lys catabolism. This seed-specific metabolic perturbation stimulated Lys accumulation starting from the initiation of storage reserve accumulation. Our results revealed that the response of seed metabolism to the inducible alteration of Lys metabolism was relatively minor, however, that which was observable operated in a modular manner. They also demonstrated that Lys metabolism is strongly associated with the operation of the TCA cycle, whilst largely disconnected from other metabolic networks. In contrast, the inducible alteration of Lys metabolism was strongly associated with gene networks, stimulating the expression of hundreds of genes controlling anabolic processes that are associated with plant performance and vigor, whilst suppressing a small number of genes associated with plant stress interactions. The most pronounced effect of the developmentally-inducible alteration of Lys metabolism was an induction of expression of a large set of genes encoding ribosomal proteins as well as genes encoding translation initiation and elongation factors, all of which are associated with protein synthesis. With respect to metabolic regulation, the inducible alteration of Lys metabolism was primarily associated with altered expression of genes belonging to networks of amino acids and sugar metabolism. The combined data are discussed within the context of network interactions both between and within metabolic and transcriptional control systems.

Experiment Overall Design: Arabidopsis thaliana (WS) seeds of the WT and KD genotype (Zhu and Galili, 2003)were germinated on soil and grown in the greenhouse (23oC). Flowers were marked and at given time intervals following flowering (14DAF to Dry ). Maturing siliques were collected, then seeds were dissected from siliques . Mature seeds were collected at the end of the desiccation period and stored at 4ºC. Two repeats were collected from each time point. Three to five thousands seeds were harvested for each extraction.

However, the molecular mechanism by which L-Ser deficiency impairs basic cellular function remains largely unexplored. We used microarrays to identify genes differentially expressed in KO-MEFs cultured with presence or absence of L-Ser, and tried to understand the underlying mechanism of growth-arrested phenotype of KO mice/MEFs elicited by L-Ser deficiency.
