Experiment Overall Design: Cultured BE(2)-C cells were differentiated with 10 uM all-trans retinoic acid (RA) for 3 weeks and infected with WEEV at an MOI=10 for 6 h, and Affymetrix Human Genome Array U133 Plus 2.0 chips were used to analyze transcript levels.
Project description:The Southern house mosquito, Culex quinquefasciatus, is a vector of the causative agents of many diseases including West Nile Fever, St. Louis Encephalitis, and lymphatic filariasis. In order to manage the spread of these diseases, vector control efforts rely heavily on insecticides, including pyrethroids, namely permethrin. In our study we investigated the changes in the gene expression profiles of a highly-permethrin resistant strain of Cx. quinquefasciatus during constant exposure to permethrin at the LC-0, LC-50, and LC-70 rates, which killed 0, 50, and 70% of all larvae, respectively. Overall, we identified several genes that were up-regulated including detoxification genes such as cytochrome P450s as well as genes that were down-regulated.
Project description:We performed whole genome single nucleotide polymorphism (SNP) based analysis of all available Venezuelan equine encephalitis (VEE) virus antigenic complex genomes and developed a high resolution genome-wide SNP microarray. We used the SNP microarray to analyze a broad panel of VEEV isolates, found excellent concordance between array and sequence based genotypes for previously sequenced strains, and genotyped unsequenced strains.

Project description:Japanese encephalitis (JE) is an acute encephalitis syndrome contributed to Japanese encephalitis virus (JEV) infection. It is the chief cause of viral encephalitis in Asian. In recent years, association of JEV infection with neurological problems such as Guillain-Barré syndrome had reported. Nevertheless, its potential pathogenic mechanism has never been reported. Therefore, it is urgent to explore the relationship between peripheral nerve injury and JEV infection. Here, we use the liquid chromatography-tandem mass spectrometry (LC-MS/MS) technique to make out the protein expression levels of mice sciatic nerve between JEV infection group and the sham group. In general, 4303 proteins were identified by MS, and 187 differentially expressed proteins were found. There were 105 proteins up-regulated in the injured sciatic nerve, and 82 proteins were down-regulated. Functional enrichment analysis of differentially expressed proteins showed that the up-regulated proteins were mainly related to immune regulatory response, and the down-regulated proteins were related to ribosomal structural components and translation.
