Project description:To assess the impact of surface water across the Hun River, several sampling sites located in the mainstream and the tributary were selected representative of pollution gradient and different pollution source. Human mesenchymal stem cells were exposed to organic extracts of surface water from six sites for 2 days. Microarrays were used to measure the gene expression. And the gene expression profiles were used to evaluate the ability of determine the potential biological effects, to differentiate different pollution source, and to identify the toxic components.

Project description:The aim of this study was to perform comparative analysis of global gene expression changes in lung BEAS-2B cells in response to treatment with extracts of particulate matter produced by conventional gasoline (E0) or biogasoline (E15) differing in the content of organic compounds, particularly PAHs. To examine the toxicity, we monitored genome-wide gene expression changes in human lung BEAS-2B cells, exposed for 4h and 24h to a subtoxic dose of each PM extract. After 4h exposure, numerous dysregulated genes and processes such as oxidative stress, lipid and steroid metabolism, PPARα signaling and immune response, were found to be common for both extract treatments. Twenty four hour exposure resulted in more distinctive gene expression patterns. Although several common modulated processes indicating the metabolism of PAHs and activation of aryl hydrocarbon receptor (AhR) were identified, E15 specifically dysregulated a variety of other genes and pathways related to cancer promotion and progression, possibly due to the increased PAH content in PM organic extract.

Project description:Here we found Rosa roxburghii fruit extracts effectively increase TERT expression and telomerase activity in cultured human mesenchymal stem cells. Both Rosa roxburghii fruit extracts by freeze drying and spray drying methods increase the activity of telomerase. Rosa roxburghii fruit freeze drying extracts is able to reduce reactive oxygen species levels, enhance SOD activity and resistance to oxidative stress, and reduce DNA damage caused by oxidative stress or radiation. Rosa roxburghii fruit extracts promoted cell proliferation, improved senescent cell morphology, delayed replicative cellular senescence, attenuated cell cycle supressors and alleviated the senescence-associated secretory phenotype. Transcriptome and metabolic profilings found that Rosa roxburghii fruit extract promote cell proliferation and DNA repair pathways, decreased triglycerides as well. Overall, we provided a theoretical basis for the application of Rosa roxburghii fruit as an anti-aging natural product.

Project description:Human mesenchymal stem cells (hMSCs) are defined as multi-potent colony-forming cells expressing a specific subset of plasma membrane markers when grown on flat tissue culture polystyrene. However, as soon as hMSCs are used for transplantation, they are exposed to a 3D environment, which can strongly impact cell physiology and influence proliferation, differentiation and metabolism. Strategies to control in vivo hMSC behavior, for instance in stem cell transplantation or cancer treatment, are skewed by the un-physiological flatness of the standard well plates. We used micrometer-scale defined surface topographies as a model to describe the phenotype of hMSCs during adaptation to their new environment. Compared to hMSCs cultured on flat polystyrene, we observed dramatically changed cell morphologies accompanied by shrinkage of cytoplasm and nucleus, a decreased overall cellular metabolism, and slower cell cycle progression resulting in a lower proliferation rate in cells exposed to surface topographies. We hypothesized that this reduction in proliferation rate effects their sensitivity to certain cancer drugs, which was confirmed by higher survival rate of hMSCs cultured on topographies exposed to paclitaxel. Thus, micro-topographies can be used as a model system to mimic the natural cell micro-environment, and be a powerful tool to optimize cell treatment in vitro. Project description:We investigated here the reprogramming of human bone marrow mesenchymal stem cells (MSC) using cell-free extracts from renal proximal tubular epithelial cells (HK2). Reprogramming of MSC is evidenced by a dramatic change in the cell structure with the formation of microvilli, the expression of specific markers and the acquisition of functional properties characteristic of the proximal tubular epithelial cells. To better characterize the MSC treated with HK2 extracts, we next isolated clones using a limiting dilution approach. Five out of 50 clones, showing the most epithelial-like morphology, were subjected to further analysis by qRT-PCR to examine their mesenchymal and epithelial marker profile. The difference in the expression of these markers reflects an heterogeneity in the reprogramming efficiency. Cells from clone 17 (CL17) were the most similar to the HK2 cells. RNA sequencing of CL17, MSC and HK2 cells was performed in order to evaluate the global effect of MSC reprogramming with cell extracts. RNA sequencing confirmed a switch of the reprogrammed cells toward a proximal tubular epithelial genotype along the EGF receptor pathway.

Project description:Arsenic and mercury are known chemical hazards. The differences in effects from organic and inorganic forms of these toxic elements is less well understood, however. The nematode Caenorhabditis elegans (C. elegans) is a suitable model to investigate the toxicity of environmental hazards. In this study, the transcriptomic profiles of C. elegans exposed to inorganic mercury chloride (HgCl2) and sodium (meta)arsenite (NaAsO2) were assessed alongside organic methylmercury chloride (meHgCl) and dimethylarsinic acid (DMA). For this purpose, adult C. elegans were exposed for 24 h to NaAsO2 (10 µg/ml), DMA (200 µg/ml), HgCl2 (2 µg/ml), and meHgCl (0.5 µg/ml), concentrations that were equitoxic in juveniles for developmental delay. Whole genome gene expression profiles were determined by using Cellegans_UnrestrictedGE_G2519F_020186 Microarray (Agilent Technologies, Santa Clara, CA). The results showed significant changes in the transcriptome of adult C. elegans exposed to NaAsO2, DMA, HgCl2, or meHgCl relative to the control group (C. elegans treated with water). A total of 927 and 1221 differentially expressed genes (DEGs) were found in C. elegans treated with 10 µg/ml NaAsO2 or 200 µg/ml DMA, respectively. Interestingly, only 161 DEGs were in common for these two chemicals. Exposure to 2 µg/ml HgCl2 or 0.5 µg/ml meHgCl altered the expression of 670 and 485 genes, respectively, and out of these genes, 154 were commonly altered by the two treatments. Analysis of DEGs revealed that organic and inorganic forms of arsenic and mercury have different effects on the transcriptome of adult C. elegans.

Project description:Genome-wide DNA methylation profiling was performed in human mesenchymal stem cells (hMSCs) differentiated into adipocytes (day 10) while being continuously exposed to either one of three different persistent organic pollutants (POPs), namely TCDD, PFOS, and TBT. The Illumina Infinium 450K Human DNA methylation Beadchip v1.2 was used to measure DNA methylation of unexposed differentiated adipocytes and compared to POP-exposed differentiated adipocytes. Our aims were to 1) measure genome-wide DNA methylation changes upon POP exposure during adipocyte differentiation in primary hMSCs and 2) investigate the relationship between DNA methylation and gene expression regulation after POP-exposure in a panel of 84 adipocyte-related genes.

Project description:Staphylococcus aureus is an important food poisoning bacterium. In food preservation, acidification is a well-known method. Permeant weak organic acids, like lactic and acetic acids, are known to be more effective against bacteria than inorganic strong acids (e.g., HCl). Growth experiments and metabolic and transcriptional analyses were used to determine the responses of a food pathogenic S. aureus strain exposed to lactic acid, acetic acid, and HCl at pH 4.5. Lactic and acetic acid stress induced a slower transcriptional response and large variations in growth patterns compared with the responses induced by HCl. In cultures acidified with lactic acid, the pH of the medium gradually increased to 7.5 during growth, while no such increase was observed for bacteria exposed to acetic acid or HCl. Staphylococcus aureus increased the pH in the medium mainly through accumulation of ammonium and the removal of acid groups, resulting in increased production of diacetyl (2,3-butanedione) and pyrazines. The results showed flexible and versatile responses of S. aureus to different types of acid stress. As measured by growth inhibition, permeant organic acid stress introduced severe stress compared with the stress caused by HCl. Cells exposed to lactic acid showed specific mechanisms of action in addition to sharing many of the mechanisms induced by HCl stress. Data is also available from http://bugs.sgul.ac.uk/E-BUGS-87

Project description:Members of the spotted fever group rickettsia express four large, surface-exposed autotransporters, at least one of which is a known virulence determinant. Autotransporter translocation to the bacterial outer surface, also known as type V secretion, involves formation of a b-barrel autotransporter domain in the periplasm that inserts into the outer membrane to form a pore through which the N-terminal passenger domain is passed and exposed on the outer surface. Two major surface antigens of Rickettsia rickettsii, are known to be exposed and the passenger domain cleaved from the autotransporter domain. A highly passaged strain of R. rickettsii, Iowa, fails to cleave these autotransporters and is avirulent. We have identified a putative peptidase, truncated in the Iowa strain, that when reconstituted into Iowa restores appropriate processing of the autotransporters as well as restoring a modest degree of virulence.
