MicroRNA differentially expressed in diapausing versus HCl-treated Bombyx embryos

Project description:Differentially expressed microRNAs (miRNAs) were detected to explore the molecular mechanisms of diapause termination. The total small RNA (sRNA) of diapause-destined silkworm eggs and HCl-treated eggs was extracted and then sequenced using HiSeq/MiSeq high-throughput method. 44 novel miRNAs were discovered. Compared to those in the diapause-destined eggs, 61 miRNAs showed significant changes in the acid-treated eggs, with 23 being up-regulated and 38 being down-regulated. The potential target genes of differentially expressed miRNAs were predicted by miRanda. Gene Ontology and KEGG pathway enrichment analysis of these potential target genes revealed that they were mainly located within cells and organelles, involved in cellular and metabolic processes, and participated in protein production, processing and transportation. Two differentially expressed genes, Bombyx mori SDH (BmSDH) and Bmo-miR-2761-3p, were further analyzed with qRT-PCR. BmSDH was significantly up-regulated in the HCl-treated eggs, while Bmo-miR-2761-3p was down-regulated. These results suggested that these two genes were well coordinated in silkworm eggs. Dual luciferase reporter (DLR) assay demonstrated that Bmo-miR-2761-3p inhibited the expression of BmSDH.

Project description:This study provides an evaluation of changes in gene expression associated with chlorpromazine HCl treatment of rat hepatocytes in vitro. Primary rat hepatocytes were treated for 24 and 48 hours with two doses (0.8 uM and 20 uM) of chlorpromazine HCl and 1% DMSO vehicle control. Five replicates of each treatment were performed. Cells were then extracted and RNA processed for microarray analysis.

Project description:Gallic acid (also known as 3,4,5-trihydroxybenzoic acid, GA), a naturally occurring phenolic acid, is a major constituent of tea and red wine. It has been demonstrated that GA possess anti-cancer activities. We found the epigenetic effect of GA in tobacco-associated human cancer cell lines. In this dataset, we include the expression array data from human lung cancer H1299 cell line with or without the treatment of 5azaC or GA. These data were used to obtain genes upregulated in both 5azaC- orGA-treated H1299 cells.

Project description:Detection, treatment, and prediction of outcome for lung cancer patients increasingly depend on a molecular understanding of tumor development and sensitivity of lung cancer to therapeutic drugs. The application of genomic technologies, such as microarray, is widely used to monitor global gene expression and has built up invaluable information and knowledge, which is essential to the discovery of new insights into the mechanisms common to cancer cells, resulting in the identification of unique, identifiable signatures and specific characteristics. It is likely that application of microarray may revolutionize many aspects of lung cancer being diagnosed, classified, and treated in the near future. Project description:To understand global expression changes in a knockdown of PGC1alpha (siPGC1alpha) vs control (siControl) in a lung metastatic cell line (4175) Metabolic adaptations play a key role in fuelling tumour growth. However, less is known regarding the metabolic changes that promote cancer progression to metastatic disease. Herein, we reveal that PGC-1a expression and activity are differentially regulated depending on breast cancer metastatic sites. Breast cancer cells that preferentially metastasize to lung display striking upregulation of PGC-1a expression. PGC-1a promotes breast cancer cell migration and invasion in vitro and augments lung metastasis in vivo. MDA-MB-231 lung variant (4175) cells were treated with a short interfering RNA (siRNA) for human PGC-1a (PPARG1A) or control siRNA for 48hrs 24 hours post-plating cells.
