CD40L modulates CD4+ T cell polarization and activation through the binding of Receptor for Activated C Kinase 1

Project description:Inhibition of the co-stimulatory ligand CD40L has shown beneficial effects in many experimental models of auto-immune disease and inflammation. Here, we show that CD40L deficiency in T-cells in mice causes a reduction of CD4+ T cell activation and specifically a strong reduction in interferon- γ (IFN-γ) producing T helper 1 (Th1) cells. In vitro, we could not reproduce this APC-dependent effects, but found that T-cell CD40L affects cell death and proliferation. We identified receptor of activated C kinase (RACK1), the canonical PKC binding partner and known to drive proliferation and apoptosis, as a mediator of CD40L reverse signaling. Furthermore, we found that CD40L clustering stabilizes IFN-γ mediated Th1 polarization through STAT1, a known binding partner of RACK1. Together this highlights the importance of both CD40L forward and reverse signaling.

The experiment shows that, as expected, cells lacking JAK2 or TNFRS1A expression display a severly blunted response to the tested cytokines. Single-cell RNA and BCR sequencing of naive B cells cultured in IgG class-switching polarising conditions

Project description:Naive B cells (CD27-IgD+) were obtained from 3 healthy controls and cultured in vitro under anti-IgM, CD40L and IFN-gamma to polarise class-switching towards IgG isotypes. Single-cell RNA and BCR sequencing libraries were prepared at Day 0 (before addition of stimuli), Day 3 and Day 6 of the cell culture time-course. This is intended as a dataset to validate a new computational method sciCSR in enumerating productive and sterile immunoglobulin transcripts as indicators of B cell class switching and maturation dynamics.

Project description:IFN-γ has been reported to be the ABC-promoting cytokine combined with signaling from IL-21, CD40L, Ag-BCR and TLR7 agonist. Hence, we adopted the in vitro differentiation cocktails (anti-IgM, R848, anti-CD40, IL-21 plus IFN-γ or IL-4) to determine the role of IFN-γ in ABC differentiation. Mouse splenic B cells were purified by negative selection and cultured in the presence of the above cocktails. Compared with IL-4, IFN-γ-polarized cells preferentially differentiated to ABC-like cells. And RNA-seq were performed on IFN-γ-polarized cells and IL-4-polarized cells.

The effects of terbutaline or GW9508 on TNF-alpha and IFN gamma (TNF-alpha + IFN gamma) stimulation by HaCaT

Project description:G-protein coupled receptors (GPCRs) have diverse roles in physiological processes, including immunity. Gs-coupled GPCRs increase while Gi-coupled ones decrease intracellular cAMP. Previous studies suggest that, in epithelial cells, Gs-coupled GPCRs enhance whereas Gi-coupled GPCRs suppress pro-inflammatory immune responses. In order to examine the issue, we chose beta2 adrenergic receptor and GPR40 as representatives of Gs- and Gi- coupled GPCRs, respectively, and examined their effects on TNF-alpha and IFN-gamma-(TNF-alpha + IFN-gamma) induced gene expression by HaCaT. We used microarrays to detail the global changes of gene expression induced by a beta2 adrenergic receptor agonist terbutaline or GPR40 agonist GW9508 pre-treatment in TNF-alpha + IFN-gamma - stimulated HaCaT cells. HaCaT cells were pre-treated with terbutaline or GW9508, TNF-alpha + IFN-gamma were then added, and cultured for another 24 h. Cells were then used for RNA extraction and hybridization on Affymetrix microarrays. We sought to clarify changes in gene expression after 1) TNF-alpha + IFN-gamma, 2) TNF-alpha + IFN-gamma + terbutaline, and 3) TNF-alpha + IFN-gamma + GW9508 treatment. To this end, we set 4 groups of samples; 1) unstimulated group, 2) TNF-alpha + IFN-gamma-stimulated group, 3) TNF-alpha + IFN-gamma + terbutaline-stimulated group, and 4) TNF-alpha + IFN-gamma + GW9508-stimulated group. In each group, HaCaT cells were stimulated in triplicate wells (n=3).
