Project description:Colorectal cancer is one of the most common cancers worldwide with increasing incidence, the presence of metastasis is one of the major causes for poor outcome. BEX2 has been reported to be involved in tumor development in several types of cancer, but is poorly understood in metastatic colorectal cancer. Here we demonstrated that knockout of BEX2 resulted in the enhancement of the migratory and metastatic potential of colorectal cancer cells in vivo and in vitro, re-expression of BEX2 in knockout cells could reverse the migratory enhancement. Expression profile chip indicated that hedgehog signaling pathway was activated after knockout of BEX2, and hedgehog Signaling inhibitor GANT61 and GDC-0449 could somehow reverse the migratory enhancement of BEX2-/- colorectal cancer cells. We also demonstrated that it is the nucleus translocation of Zic2 after BEX2 silenced, that activated hedgehog signaling pathway, while knockdown Zic2 could also abrogated migratory enhancement of BEX2-/- cells. In summary, our findings suggest that BEX2 is a negative modulator of hedgehog signaling pathway by retaining Zic2 in the cytoplasm of colorectal cancer cells, thus to inhibit colorectal cancer cell migration and metastasis.

Project description:Colorectal cancer is a common cancer and causes high mortality worldwide. It is urgent to explore its pathogenesis and seek effective therapeutic strategies. Here, the author showed that EEF1D expression in colorectal cancer cells is required to promote tumor growth and metastasis. Moreover, we further performed the proteomic analysis to gain a deeper mechanistic understanding of EEF1D in CRC. We found that EEF1D interacted with various cancer-related proteins. These interactions further support the importance of EEF1D in colorectal cancer.

Project description:Liver metastasis is one of the major causes of death in colorectal cancer (CRC) patients. To understand this process, we investigated whether the gene expression profiling of matched colorectal carcinomas and liver metastases could reveal key molecular events involved in tumor progression and metastasis. We performed experiments using a cDNA microarray containing 17,104 genes with the following tissue samples: paired tissues of 25 normal colorectal mucosa, 27 primary colorectal tumors, 13 normal liver and 27 liver metastasis, and 20 primary colorectal tumors without liver metastasis. To remove the effect of normal cell contamination, we selected 4,583 organ-specific genes with a false discovery rate (FDR) of 0.0067% by comparing normal colon and liver tissues using significant analysis of microarray, and these genes were excluded from further analysis. We then identified and validated 46 liver metastasis-specific genes with an accuracy of 83.3% by comparing the expression of paired primary colorectal tumors and liver metastases using prediction analysis of microarray. The 46 selected genes contained several known oncogenes and 2 ESTs. To confirm that the results correlated with the microarray expression patterns, we performed RT-PCR with WNT5A and carbonic anhydrase II. Additionally, we observed that 21 of the 46 genes were differentially expressed (FDR = 2.27%) in primary tumors with synchronous liver metastasis compared with primary tumors without liver metastasis. We scanned the human genome using a cDNA microarray and identified 46 genes that may play an important role in the progression of liver metastasis in CRC. Project description:Metastasis is the major cause of cancer-related death in patients with colorectal cancer. Although inducible nitric oxide synthase (iNOS) is a crucial regulator of cancer development and progression, its roles in epithelial-mesenchymal transition (EMT) and the pathogenesis of metastatic colorectal cancer have not been fully investigated. Primary colorectal cancer and liver metastatic tissue specimens were analyzed showing 90% of liver metastatic colorectal cancer with reduced expressions of iNOS compared with 6% of primary colorectal cancer. The Cancer Genome Atlas database analyses via cBioPortal reveal that mRNA expression of iNOS negatively correlated with selected EMT markers in colorectal cancer in a cancer type-dependent manner. The transcriptomic profiling (RNA sequencing data) indicates that iNOS knockdown in SW480 colorectal cancer cells induced an EMT program with upregulated expression of selected stem-cell markers. iNOS knockdown did not alter E-cadherin mRNA expression but re-localized it from membrane to cytoplasm through iNOS-GATA4-Crb2-E-cadherin pathway. iNOS knockdown induced a change in cell morphology, and promoted cell invasion and migration in vitro, and metastasis in vivo.

Project description:The study identifies a set of genes through functional selection in an orthotopic colorectal cancer mouse model that play an important role in mediating colorectal cancer liver metastasis. We look at the gene profiling of metastatic colorectal cancer cells (L1 and L2) and non-mestastatic colorectal cancer cells (sw620 and sw480) using Affymetrix U133A oligonucleotide arrays. 4 samples (each with triplicates) were analyzed. Project description:The survival rate of colorectal liver metastases remains low. Long noncoding lncRNAs is emerging as a novel class of master regulators of cell invasion and metastasis. In this study, analysis of the lncRNA expression dynamics in the colorectal liver metastases, primary colorectal tumor and normal tissues led to the identification of the metastasis-specific expression of a set of lncRNAs including CRLM1. CRLM1 inhibited apoptosis and promoted metastasis and invasion of colorectal cancer cells, and also promoted tumor growth in nude mice. CRLM1 weakly associated with chromatin regions of genes involved in cell adhesion and DNA damage, correlated with CRLM1-regulated gene expression bidirectionally. CRLM1 physically interacts with and promotes the nuclear localization of the metastasis protein hnRNPK. CRLM1 effectively promotes hnRNPK promoter occupancy, and co-regulate gene expression. These findings suggest a potential biomarker and druggable target for treatment of colorectal liver metastases.
