Project description:The current methods to distinguish slow-growing from aggressive localised prostate cancer at diagnosis are imprecise so aggressive tumours can sometimes be missed. In this study we examined whether the changes in the features of cancer associated fibroblasts (CAFs) from the surrounding tumour microenvironment can help identify high risk tumours. We show that CAFs have a distinct methylation profile versus non-malignant prostate fibroblasts (NPFs) including specific differentially methylated regions that distinguish higher grade prostate cancer.

Project description:We aimed to investigate genome wide DNA methylation changes in human colon cancer organoids with the BRAF-V600E mutation and in human colon cancer organoids in which the BRAF-V600E mutation was corrected by means of CRISPR genome editing. DNA methylation analysis was performed using the Infinium Methylation EPIC beadchip at the Pathology department of the UMC Utrecht (The Netherlands).

EPIC-Italy at HuGeF

Project description:This Series contains data from 845 participants (188 men and 657 women) in the EPIC-Italy cohort that was produced at the Human Genetics Foundation (HuGeF) in Turin, Italy. At the last follow-up (2010), 424 participants remained cancer-free, 235 had developed primary breast cancer, 166 had developed primary colorectal cancer, and 20 had developed other primary cancers. Anthropometric measurements, and dietary and lifestyle information obtained by questionnaire are also available. A total of 845 samples from the EPIC-Italy cohort were analyzed.

Identification of the molecular signature of distinctive breast cancer associated fibroblasts (CAFs)

Project description:Cancer associated fibroblasts (CAFs) are highly heterogeneous and different subsets of CAFs may exhibit distinct functions, To identify the molecular signature of distinctive CAFs , we compared mRNA expression profiles of CAFs isolated from tumors in sensitive patients and resistant ones before neo-adjuvant chemotherapy. Compared the CAFs from sensitive samples, those from refractory samples exhibited a distinctive signature.
