DNA methylation profiles of F1 mice sperm of the control group and arsenic group

Project description:Pregnant C3H mice were given tap water (control group) and tap water containing 85 ppm sodium arsenite from gestational day 8 to 18 (arsenic group), respectively. The DNA methylomes of sperm of F1 mice were investigated by RRBS method. The results showed that gestational arsenic exposure increased hypomethylated cytosines in the F1 sperm genome. The present study has indicated environmental impacts on sperm DNA methylome establishment.

Project description:Ground water Arsenic toxicity is the global problem and millions of people are exposed to elevated levels through drinking water than WHO permiscible limit of 10µg/L. The exposure is associated with various cancerous and non-cancerous diseases. It may alter the gene expression profile of invidual and suppress the activity of various genes giving rise to different diseases. Pakistan, a developing country in South Asian region, also have different areas reported to have elevated Ground water As. levels since 2005. The Present Study aimed to investigate Transcriptome profile of the exposed subjects in two districts of Punjab Province Pakistan. i.e Lahore and Kasur. The population was stratified into three exposure groups comprising Low, Medium and High exposure based on their urinary arsenic levels. Fundulus exposed to arsenic as embryos

Project description:The present study used microarray expression profiling to determine the effects of embryonic arsenic exposure. Fertilized killifish (Fundulus heteroclitus) eggs were exposed to 0, 5, 15, or 25ppm arsenic as sodium arsenite. To examine differentially expressed genes, the microarrays were probed using RNA obtained from the control and 25ppm-exposed killifish just after hatching. No differences were noted in survival or hatching success between any of the groups. After analysis, a set of 332 genes was found to accurately distinguish between the control and 25ppm exposure groups. Expression of several of the genes (CDBP1, Arts1, FetB, and Fbp7) was quantified by qPCR in the lower exposure groups and at earlier time points to examine temporal and dose-responsive expression patterns. These results will enable us to better understand how arsenic impacts development. Killifish eggs were fertilized, divided into petri dishes containing 40 eggs (n=10 replicate petri dishes), and cultured until hatch in 0 or 25 ppm arsenic as sodium arsenite. Four to five hatchlings within each petri dish were pooled to obtain RNA. A total of 20 arrays were probed, 10 with RNA from control fish and 10 with RNA from the arsenic-exposed fish.

Project description:In utero exposure to arsenic via drinking water increases the risk of lower respiratory tract infections during infancy and mortality from bronchiectasis in adulthood. Pregnant mice were exposed to arsenic, and gene expression patterns were profiled in peripheral lung tissue obtained from the offspring at 2 weeks of age. Pregnant Mice (BALB/c, C57BL/6, C3H/HeARC) mice were exposed to arsenic (or control) via drinking water from day 8 of gestation until the birth of their offspring. After giving birth, mothers were given control drinking water. At 2 weeks post-natal age, total RNA was extracted from peripheral lung tissue of the offspring and analysed on Affymetrix microarrays.

The effects of arsenic on Silurana tropicalis embryos

Project description:Arsenic is a carcinogenic metalloid element found naturally in the environment; however human activities such as mining and pesticide use have led to increased concentrations in some areas. Silurana tropicalis eggs were exposed to sublethal concentrations of sodium arsenate (0, 0.5 and 1 ppm) for 3 days during embryogenesis. Four pooled of 10 larvae were sampled per treatment and analyzed for gene expression changes using a microarray approach. Silurana tropicalis eggs were exposed to sublethal concentrations of sodium arsenate (0, 0.5 and 1 ppm) for 3 days during embryogenesis. There were 5 controls, 5 low dose and 6 high dose microarrays perfomed.
