Project description:A Saccharomyces cerevisiae mutant with improved coniferyl aldehyde resistance was obtained by using an evolutionary engineering strategy, based on successive batch cultivation under gradually increased coniferyl aldehyde concentration. The mutant strain BH13 was selected which could grow at a coniferyl aldehyde concentration that the reference strain could not grow at all. Whole-genome transcriptomic analysis of BH13 with respect to the reference strain was performed to identify differences in gene expression levels between the two strains.

Transcriptional response of Saccharomyces cerevisiae to the proapoptotic unsaturated fatty aldehyde t-2-hexadecenal by RNAseq

Project description:Hexadecenal, a trans (-2,3-) unsaturated fatty aldehyde, is an intermediate of the sphingolipid degradation pathway. This pathway, induced during cellular stress, involves the conversion of sphingosine-1-phosphate to hexadecenal by Sphingosine-1-Phosphate-Lyase (SPL) in humans and DPL1 in yeast. Hexadecenal is further metabolized to hexadecenoic acid by Fatty Aldehyde Dehydrogenase (ALDH3A2 in humans and HFD1 in yeast). Trans-2-hexadecenal (t-2-hex), has been found to induce mitochondrial dysfunction in a conserved manner from yeast to humans. However, the specific mechanisms and biological targets underlying this lipid-induced mitochondrial inhibition remain largely unknown. In this study, we employed unbiased transcriptomic approaches using the Saccharomyces cerevisiae yeast model to elucidate the principal mechanisms and biological targets associated with t-2-hex-induced mitochondrial dysfunction. To investigate the effects of t-2-hex, we utilized an hfd1 mutant strain lacking the HFD1 gene. This strain exhibited increased sensitivity to t-2-hex treatment compared to wild-type cells. Exponentially growing hfd1 mutant cells were exposed to 100 μM t-2-hex for a duration of 3 hours, while control cells were incubated with the solvent dimethyl sulfoxide (DMSO), in which hexadecenal is dissolved. Total RNA was extracted from both treated and control cells for high-throughput sequencing analysis. Through transcriptomic profiling, we aimed to identify differentially expressed genes, pathways, and regulatory networks associated with t-2-hex-induced mitochondrial dysfunction in yeast. This study provides a comprehensive analysis of the transcriptional response to t-2-hex treatment, shedding light on the molecular mechanisms and potential biological targets underlying lipid-induced mitochondrial dysfunction.

Project description:Iron-resistant Saccharomyces cerevisiae mutant was obtained by evolutionary engineering selection strategy. The mutant obtained M-bM-^@M-^\M8FEM-bM-^@M-^] is much more resistant to iron stress than the reference strain which was used to select this mutant. Mutant can resist up to 35mM Iron* stress whereas the reference strain cannot. Briefly, genetic diversity in reference strain, CEN.PK.113-7D, was increased by ethyl methane sulfonate (EMS)-mutagenesis. The mutant population was passaged several times in gradually increasing silver stress. Several mutant individuals were selected from the final population. Among selected mutant individuals, one of them was much more resistant to silver stress than the reference strain, called as 2E. Whole-genome transcriptomic analysis was performed to identify the silver resistance mechanisms in the silver-resistant mutant strain.

Project description:NaCl-resistant Saccharomyces cerevisiae mutant was obtained by evolutionary engineering. EMS mutagenized culture was used as the initial population for the selection procedure. Gradually increasing levels of NaCl stress was applied through 40 successive batch cultivations. The reference strain could not grow even at 0.85 M NaCl whereas this mutant was shown to be resistant up to 1.45 M NaCl concentration. Whole-genome microarray analysis was used to identify the NaCl resistance mechanisms by comparing NaCl-resistant mutant strain and the reference strain in the absence of NaCl stress. The reference Saccharomyces cerevisiae strain and the NaCl-resistant mutant were grown in minimal medium to an Optical Density (OD600) of 1.00 which corresponds to the logarithmic growth phase of the yeast cells. Cultures were harvested and whole RNA isolation was carried out. The experiment was repeated three times.
